Purpose:To investigate the correlation of Shh and Ptch, which are two core protein of Hedgehog pathway, expression with clinicopathological characteristics and tumor recurrence after radical surgery resection of Hilar cholangiocarcinoma and analyze molecular mechanisms of invasion and recurrence after radical surgery resection in hilar cholangiocarcinoma.Methods:1. Immunohistochemistry was applied to detect the expression of Shh and Ptch expression in 44 specimens of hilar cholangiocarcinoma and 7 specimens of benign bile duct tissues. The relation of the expression to clinicopathological characteristics and tumor recurrence after radical resection was analyzed.2. A multivariate regression analysis was performed and be tested by Log-rank test. Selecting some reflect factors in these patients and analyzing by the computer's Cox proportional hazard model.Results: The expression intensity of Shh and Ptch, which are two core protein of Hedgehog pathway, in Hilar cholangiocarcinoma was remarkably stronger than that in benign bile duct tissue; Correlation analysis revealed that the expression of Shh and Ptch was positively related to the progress of the tumor invasion and tumor recurrence after radical surgery resection; The univariate statistics shows the more higher Shh and Ptch expression in Hilar cholangiocarcinoma the more shorter tumor recurrence after radical surgery resection (P<0.01). Multivariate analysis revealed that the expression of Shh and Ptch was the most important predict factor for tumor recurrence after radical surgery resection of Hilar cholangiocarcinoma (P<0.01).Conclusion:The expression of Shh and Ptch was related to the progress of the tumor invasion and tumor recurrence after radical surgery resection.
