Objective: Restraint the expression of Ppif gene, observation the protective effect of this method for renal ischemia-reperfusion injury in rats, and Analysis, discussion of its mechanism.Methods: First, to made a renal ischemia-reperfusion model in rats, and then they were evenly divided into three groups: control group, negative control group, and treated group(20 rats in each group). When the IRI was occur, RNAi lentiviral vector of Ppif gene targeting was given to treated group, dose of 0.3 mL(4μg/ g), and normal saline(with volume fraction of 0.01 DMSO), 0.3 mL, to NC group; The CON group wasn't clamped the renal pedicles, that the renal didn't encounter the IRI. Scheduled time comes, to find the result of the blood urea nitrogen (BUN) and creatinine (Cr), to count the renal cell apoptosis index (AI), to observe the histopathological analysis of HE staining.Results: The result showed the levels of Cr and BUN were both decreased between treated group and CON group, the AI decreased, (P < 0.05), there are Statistically significant difference between them. Pathological results(HE staining): A comparison between 3 group rats, the ischemia of the treated group significantly reduced than other two ones.Conclusions: The expression of Ppif Gene of rats can be restrain effectively by RNAi lentiviral vector of Ppif gene targeting, that we can reduce apoptosis of Mitochondrial pathway. Finally, when the IRI was occur, the kidney can be protected.
