Objective: We hypothesized that IL-17 displays potentially critical role in host defense agaist Pseudomonas aeruginosa.Methods: 1. Concentration of IL-17, IL-1β,G-CSF and MIP-2 was measured after challenging with Pseudomonas Aeruginosa at 24h, 48h and 72h.2. Exogenous mIL-17 protein was pre-administered IN 12h before. After challenging with Pseudomonas Aeruginosa 6h later, concentration of IL-17, IL-1β,G-CSF, MIP-2 and bacterial clearance and survival was measured.Results: 1. Concentration of IL-17, IL-1β,G-CSF and MIP-2 increased after challenging with Pseudomonas Aeruginosa at 24h, 48h and 72h. And neutrophil cells recruitment augmented.2. Pre-administered exogenous mIL-17 protein enhanced bacterial clearance and survival after challenge with Pseudomonas Aeruginosa. IL-17, IL-1β,G-CSF, MIP-2 and neutrophil cells also increased.Conclusion: IL-17 is critical molecules expressed in response to invading pathogens and is necessary for normal lung bacterial host defense.
