Objective Investigate the role of fibronectin-integrin in the regulation of the secretion function of asthmatic airway smooth muscle cells. Methods Primary ASMCs were cultured by digestion and tissue adherent. ASMCs were be identified by light microscopy and immunohistochemistry; ASMCs or asthmatic ASMCs were inoculated on the blank plates or on the plates coated with FN, cells pretreated with integrin antibody for 30 min, non-integrin antibody treated ASMCs as control. The expressions of RANTES,TGF-β1 mRNA were observed by RT-PCR and RANTES protein in the culture supernatants was detected by enzyme-linked immunosorbent assay(ELISA). The expression of focal adhesion protein in the cell climbing film was disclosed by immunohistochemistry. Results Under the light microscope, ASMCs of tracheal or bronchial origin appear flattened and ribbon or spindle-shaped, the expression of smooth muscle actin dispersed the cytoplasm, consistent with the morphological and biological characteristics of smooth muscle cells. The expressions of RANTES,TGF-β1 mRNA of ASMCs and the RANTES protein in the ASMCs culture supernatants treated with FN were significantly increased than those in the asthmatic ASMCs without treatmen(tP<0.05); compared with those in the asthmatic ASMCs only treated with FN and without treatment,the expressions of RANTES ,TGF-β1 mRNA of ASMCs and the RANTES protein in the ASMCs culture supernatants were significantly lower in the asthmatic ASMCs treated with integrin antibody (P<0.05). Conclusion These results suggest the integrin may play an important role in the secretion function of the extracellular matrixes to the asthmatic airway smooth muscle cells.
