The Protective And Therapeutic Effect Of Dietary Nucleic Acid (zhen Ao Gan Tai) On Alcohol Liver Disease In Rats

Objective:To investigate the Protective and Therapeutic effect of dietary nucleic acid(Zhen ao Gan tai) on alcohol liver disease in rats。. Methods:Seventy-two male SD rats were separated randomly into four groups: normal control group (10), nucleic acid preventive group(12), nucleic acid treated group(12) and model group(38), the model group was separated model group one(18 rats were fed for 4 month) and model group two(20 rats were fed for 6 month), rats were fed a alcohol liquid diet and nucleic acid individually by intragastric administration except the rats of normal control group. Serum liver function(ALT, AST), blood lipids(T-CHOL, TG), MDA, SOD, GSH, PC-Ⅲ, HA andⅣ-C in the serum were measured. Light and electron microscope were adopted to observe the histopathology of the hepatic tissue. Masson dyeing was evaluated to reflect the level of hepatic fibrosis. Meanwhile, we measure the expression ofα-SMA in liver tissue by immunohistochemistry.Results:Compared with the model group, it revealed that nucleic acid preventive group and nucleic acid treated group rats:①Liver function(ALT) was obviously improved.②The level of serum cholesterin was obviously decreased.③The activity of serum SOD was increased.④The levels of serum HA was decrease.⑤Histopathological analysis of HE dyeing and electron microscope revealed that nucleic acid preventive group and nucleic acid treated group were improved.⑥Masson dyeing revealed that liver collagen fibers was markedly decreased in hepatic tissue, the degree of fibrosis was improved.⑦the expression ofα-SMA were significantly reduced compared with the model group, which indicated hepatic injury of ALD were alleviated.Conclusion:The study of the protective and herapeutic effect of dietary nucleic acid (Zhen ao Gan tai) on alcohol liver disease in rats demonstrates that dietary nucleic acid(Zhen ao Gan tai) can prevent effectively and significantly alleviate the alcohol-induced liver injury in rats, and has protective and therapeutic effect on alcohol fatty liver, hepatic fibrosis.
