Objective: Cycloxygenase is the key enzyme of prostaglandin synthesis from arachidonic acid, taking great part in the progress of Inflammation. Cycloxygenase consists of three isoforms:COX-1,COX-2 and COX-3. Domestic and foreign research have found that COX-2 overexpressed in many tumor issues as colon carcinoma, and was closed relevant with tumorigenesis and development of carcinomer. Celecoxib is of the second generation of Cycloxygenase-2 inhibitor. We try to study the influence of celecoxib on the growth of HepG2 in vitro.Methods: (1)HepG2 was cultured in the environment of 37oC,5%CO2. At the logarithmic growth phase, the cell line was treated by 5 kinds of concentration gradient celecoxib as 6.25,12.5,25,50,100μmol/l.(2)Making the culture solution of no drugs as control, and HepG2 cell line treated by the same concentration of solvents as blank, measure the OD value at 570nm through the methods of MTT after 24,48 and 72 hours'treatment.Calculate the inhibition rates and mapping according to the formula.(3)Making the normal cell line HepG2 as control, evaluate the apoptosis rate after 24h treatment by 5 different kinds of concentration of celecoxib through flow cytometry AnnexinV/PI. Analysis the results by software ModFitLT.(4)Making the normal cell line HepG2 as control, assess the cell cycle after 24h treatment by 5 different kinds of concentration of celecoxib through flow cytometry PI. Analysis the results by software ModFitLT. Results: (1)It reveals that the 6.25μmol/l celecoxib solution can't inhibit the growth of HepG2 after 24 and 48 hours'treatment(p>0.05), but plays great role at 72h(p<0.05).The other 4 concentration drugs can markedly attenuate the growth of tumor at 24,48 and 72h(p<0.01).And the ability of inhibition of celecoxib depends on the concentration and time. (2)Every group can prompt the apoptosis of HepG2 after 24h'treatment.Compared with the control, the p value is below 0.05. (3)The cells at GO/G1 phase are more than others after 24h'treatment by celecoxib compared with the control(p<0.05).At the same concentration, the difference of each phase exists statistically significance(p<0.05).Conclusions: (1)Celecoxib can significantly inhibit the growth of hepatic carcinoma cell line HepG2 cultured in vitro. (2)The ability of inhibition depends on the concentration and time of celecoxib. (3)Celecoxib can promote the apoptosis of HepG2 and prevent the cell cycle at G0/G1 phase.
