| Objective To investigate the relationship of expression of IL-17 in cardiac muscles and heart failure in rats with chronic heart failure.Methods Abdominal aorta was constricted to induce chronic heart failure in rats, echocardiogram was used to test whether the heart failure models were achieved. Rats were divided into 4 groups:sham group,heart failure group,IgG group and Ab group.The first day after operation, PBS solutions were intraperitoneally injected to sham group and heart failure group, mouse isotype IgG1 and PBS to IgG group, monoclonal human IL-17 antibody and PBS to Ab group, 5 times every two days.4 weeds,8 weeks and 16 weeks After operation, echocardiogram was used to measure and record heart failure related parameters, immuno- histochemistry method,laser scanning confocal microscope and Western Blot were used to investigate expression of IL-17 in cardic muscles.Results (1)Echocardiogram demonstrated that rat models were successfully made.(2)Compared to sham group,expression of IL-17 in cardiac muscles in heart failure group,IgG group and Ab group increased obviously(P<0.05),heart failure related parameters LVEDD,LVESD,EDV,ESV,LV/BW increased(P<0.05),LVEF,LVFS decreased(P<0.05); Compared to heart failure group and IgG group, expression of IL-17 in Ab group was lower(P<0.05),and the parameters improved (P<0.05). While,there were not obvious differences between heart failure group and IgG group.(3)Statistic analysis showed that there was a positive linear correlation between expression of IL-17 and LV/BW(r=0.880,P<0.05),a negative linear correlation between expression of IL-17 and LVEF(r=-0.987,P<0.005). Conclusion After constriction of abdominal aorta,expression of IL-17 in cardiac muscles increasd obviously,and heart failure related parameters aggravated simultaneously.Pretreated with monoclonal IL-17 antibody, expression of IL-17 decreased and parameters improved.Therefore,our study show that massive expression of IL-17 is intensively related to the development of chronic heart failure. Programmed electrical stimulus was imposed on the rat hearts of the four groups to induce ventricular arrhythmia (ventricular tachycardia and ventricular fibrillation),the evoked electrocardiograms and evoked rates were recorded. Expression of IL-17 in cardiac muscles was tested by Western Blot.Result (1)Echocardiogram showed that heart failure was induced successfully. (2) Compared to sham group,expression of IL-17 in cardiac muscles in heart failure group,IgG group and Ab group increased obviously (P<0.05) at 4 weeks,8 weeks and 16 weeks after operation,also increased the evoked rates of ventricular arrhythmia(P<0.05); Compared to heart failure group and IgG group, expression of IL-17 in Ab group was lower(P<0.05),and the evoked rates decreased(P<0.05)too.While, obvious differences were not found between heart failure group and IgG group.(3) Statistic analysis showed that there was a positive linear correlation between expression of IL-17 and the evoked rate(r=+0.954,P<0.05).Conclusion The expression level of IL-17 is perhaps related to the happening of ventricular arrhythmia in chronic heart failure rats. |
