Skin which is on body surface protects the internal tissues from injury to the external stimuli and reflects the general situation as a window. Skin abnormalities or diseases not only affect people's normal living, even worse threat to human life. Optical methods play an important role in the skin disease non-invasive detection. The current diagnostic methods appear many deficiencies, such as the optical spectral technique of a lack of structure information, images detection of lack of composition information, hyperspectral detection of a lack of the interactive information of structure and composition, and so on. Thus, the Virtual Internal Hyperspectrum of Body Surface (abbr. VIHBS) was proposed in our group base on the"banana"theory that described the photons average path in tissue. The technology was developed for obtaining the spectral and image information to reflect the changes in the composition and structure by constructing the relation curve of position, wavelength and light intensity. Besides, in order to eliminate the ambient light influence and inhibit the dark current, the modulation technique was introduced to the VIHBS detection.The VIHBS system was built to the tissue detection by VIHBS. The system that accomplishes the data collection and storage contains a modulation light, an auto translational stage, spectrometers and a personal computer. The modulation light providing the input modulated signal to the experiment system contains design of mechanical optical chopper and serial port communication with master computer. The auto translational stage design contains two parts, mechanical structure and control system, to make the fiber detector do the controlled to-and-fro movement in the linear range. The data collection software of master computer was developed by the LabVIEW language. It ensured the communication between the master computer and slave computer and completed the data collection. And then the stability test of the VIHBS system was done.The research mainly included the feasibility experiment, the validity problem of the modulation technique and the preliminary application of VIHBS. In the feasibility experiment, the comparison of VIHBS feature for normal apple and artificially doped apple was conducted to classification and location the abnormal species. In addition, the experiment proved that the VIHBS was better than reflectance spectra in the classification. The modulation test showed that the modulation technique could eliminate the ambient light influence and inhibit the dark current of instrument by comparing the modulated and un-modulated VIHBS maps in the artificial interference environment. Finally, a preliminary study was preformed for VIHBS using the modulated light to the subjects of pork. The simulation experiment was employed to make the pork changes of structure and composition. The results show that the VIHBS can achieve the location of foreign body (red filter) by visible-light and screen rapidly for changes in tissue composition caused of Intralipid by near infrared light.With further research, it will be likely to achieve the VIHBS information procession simultaneously, which use the statistical or data mining tools to integrate VIHBS data and one or more characteristics of tissue. It might provide a new method to realize the early diagnosis of skin changes in structure and composition, and might be important in various clinical applicatious.
