Objective:To tentatively explore the mechanism of HBV invading into human sperm through detecting the expression of the asialoglycoprotein receptor on human sperm,to provide new ideas and theoretical basis for blocking the paternal fetal transmission of hepatitis B virus.Methods:The semen sample were obtained from 5 cases with serum HbsAg(+)/HbeAg(+)(double- positive group),5 cases with serum HbsAg(+)/HbeAg(-)(single- positive group)and 5 cases with serum HbsAg(-)(negative group. After the plasma membrane and the cytoplasm had been isolated from the processed sperms with Plasma Membrane Protein Extraction Kit,the expression of the asialoglycoprotein receptor were detected with ASGP-R Monoclonal antibodies by Western blot. The semen sample from 11 cases with serum HbsAg(+)/HbeAg(+),23 cases with serum HbsAg(+)/HbeAg(-)and 30 cases with serum HbsAg(-)were randomly selected,then positive rate of the processed sperm were measured with ASGP-R Monoclonal antibodies by indirect immunofluorescence markings and Flow Cytometry.Results:ASGP-R existed in the plasma membrane and the cytoplasm of human sperm. The expression level of ASGP-R in negative group,single- positive group and double- positive group increased gradually. It had significantly higher positive rate of ASGP-R in single- positive group and double- positive group than those in negative group,but there was no significant differences between double- positive group and single- positive group.Conclusion:There was ASGPR in the plasma membrane and the cytoplasm of human sperm. HBV may invade into the human sperm by ASGP-R which exist in the plasma membrane and HBV may increase the expression level of ASGP-R by means of some mechanism.
