Object: To detect microRNA expression profile of human endometrium during the implantation window, and establish a biological information network involving miRNAs regulation in the process of embryo implantation.Methods: Double channel microarray technology was applied to detect miRNA expression profile of human endometrium during the implantation window, and MMIA online application server was used for target mRNA gene prediction, then building a biological information network which composed of miRNAs and mRNAs using Cytoscape software, finally MAS system was apllied for function classification of differentially expressed mRNA genes in the above network, and further forecasting some key molecular pathways in the process of embryo implantation involving miRNA regulation.Results: 1. There were 72 miRNAs differentially expressed during the implantation window, including 50 up-regulated miRNAs and 22 down-regulated miRNAs (fold change =2, q value< 0.01); 2. Excepting 23 miRNAs, the remaining 49 miRNAs were successfully forecast 6106 predicted target mRNAs genes which have a unique official gene symbols, and there were difference between the numbers of target genes of each miRNA. 3. 218 differentially expressed miRNA and mRNA gene were screened out to establish a biological information network, and there were 39 miRNAs and 114 mRNAs up-regulated, 6 miRNAs and 59 mRNAs down-regulated in the network (fold change =2.5, p value< 0.01).Conclusions: Microarray technology and computer methods could effectively analyze gene expression during the implantation window, and provide highlighted reference for the research of molecular regulation mechanism in the process of embryo implantation.
