Object: This study detected PLCE1 expression situation in normal gastric mucosa tissue and gastric cancer tissue by immunohistochemistry SP method, and research the connection between it and the clinical and Pathological features of gastric cancer, discuss the correlation between PLCE1 expression in gastric cancer and the occurrence,the development, the biological characteristics and the prognosis of gastric cancer, and lay the foundation for further studying the function of the PLCE1 gene and the role in the tumor forming process, and to provide a theoretical basis and experimental foundation for early diagnosis, prognosis and future PLCE1 target gene therapy.Method: Select 156 cases with gastric cancer tissue by surgery and pathologically confirmed which comes from General Surgery, Union Hospital, Wuhan surgery from April 2008 to September 2010 and surgical specimens of gastric cancer cutting edge of the 123 cases of gastric mucosa tissue,preoperative line Any chemotherapy and radiotherapy, and biopsy over the age: 32-80 years age group of gastric cancer, with an average age of 56, at 1997 International Union Against Cancer (UICC) TNM staging of gastric cancer reported: I of 12 cases,Ⅱstage 16 cases,Ⅲof 68 cases,Ⅳin 60 cases, according to histological grade: Well differentiated: 21 cases, moderately differentiated: 54 cases of poorly differentiated: 81 cases. This study detected PLCE1 expression in tissue by immunohistochemistry SP method to analyze its expression level and to explore the correlation between PLCE1 expression and different ages, gender, degree of differentiation, clinical stage of gastric cancer.Result: Immunohistochemistry showed that PLCE1 was mainly expressed mainly in the cell membrane, and very little in nucleus. The positive cells were seen under a microscope with brown cell membrane.PLCE1 was negative expressed or low expressed in normal gastric tissue(44.0%); in gastric cancer tissues,PLCE1 expression was higher(87.2%), the difference was statistically significant(P<0.01).Experimental results show that with the degree of increase in clinical stage, PLCE1 expression showed an increasing trend, the difference was statistically significant (P <0.01).It also proved that PLCE1 expression levels decrease with the increase in the degree of differentiation in the different histological differentiation of gastric cancer specimens, the difference was statistically significant (P <0.05).Conclusion: PLCE1 expression in gastric cancer tissues was significantly higher than that of normal gastric tissues; this suggesting PLCE1 overexpression may play an important role in the occurrence and development of gastric cancer. With study of clinical and pathological study of gastric cancer we found, PLCE1 expression decreased with the degree of tumor differentiation increased, this suggesting PLCE1 may induce tumor cell differentiation; but the study also found that there is a correlation between PLCE1 protein expression and tumor invasion depth, TNM staging and Lymph node metastasis, this suggesting PLCE1 plays an important role in the development process of the tumor, and indicating that the expression intensity of PLCE1 has a significantly effect on the degree of tumor differentiation and development trends, and to a certain extent, suggest the prognosis of gastric cancer. Detecting of PLCE1 expression contributes to infer the degree of malignancy of gastric cancer, help to assess the prognosis of patients with gastric cancer and treatment.
