Objective1)To explore the expression of TrkB in human Malignant Melanoma and investigate the role of TrkB in Malignant Melanoma.2)To research the expression of TrkB in human Malignant Melanoma cells A875, and explore the changes after using Triptolide( a kind of chemotherapeutic drugs) to recognize the possible antineoplastic mechanism of Triptolide.Methods1)The expression of TrkB in group A (40 specimens of Melignant Melanoma) and group B (30 specimens of nevus) were detected by immunohistochemical staining.2)To test the expression of TrkB in A875 cells by cellular immunohistochemisty, RT-PCR and western-blot before and after the interference with Triptolide.Results1)The positive rate of TrkB in Malignant Melanoma was 77.5% while the positive rate in nevus was 16.7%. Its expression was higher in Malignant Melanoma than in nevus, with a significant difference (P<0.01).2)a〉As a result of the cellular immunohistochemisty, the positive rate of TrkB in A875 cells after the interference was lower than cells without interference. There was an apparent difference between them (P<0.05). b〉western-blot revealed the expression after the interference of Triptolide was manifestly lower than those cells without interference(P<0.05). c〉RT-PCR displayed that the expression with the interference of Triptolide was apparently lower than cells without interference(P<0.05).ConclusionsThe expression of TrkB was positive both in Malignant Melanoma and the A875 cells. Studying of the A875 cells, we found Triptolide can inhibit the expression of TrkB, implying this may be the possible antineoplastic mechanism of Triptolide.
