Psoriasis is a common chronic inflammatory skin disease, and hypoxia is a common feature of the inflamed epidermal environment. however, little is known about the pathogenesis of the upregulation of macrophage inflammatory protein-3/CCL20 under the condition of hypoxia. We report here that CCL20 is markedly upregulated in peripheral blood of psoriasis patients comparing to the normal individuals, We also found that CCL20 expression was positively associated with HIF-1α(r = 0.855, p<0.05) by using RT-PCR,and ELISA. The levels of CCL20 in keratinocytes became higher after hypoxia. And both HIF-1αand CCL20 would be significantly downregulated after transfection of HIF-1αsiRNA into hypoxic-cultured cells . Taken together, these findings suggest that CCL20 may play a role in the immunopathogenesis of psoriasis and HIF-1αmight contribute CCL20 upregulation in psoriasis.
