Objective:The goal of this study was to evaluate the effects of berberine on the inhibition of vascular SMC proliferation and carotid artery balloon-injury model rat neointimal formation and restenosis,and explore whether the mechanisms by changing the expression of PTEN achieved.Methods: Application of MTT colorimetric assay to detect the effect of different concentrations of berberine on vascular smooth cell proliferation, RealTime-PCR and Western blot analysis showed that the effects of berberine on the vascular SMC PTENmRNA and PTEN expression at the protein level. Rat carotid artery balloon-injury models were used to investigate the effect of berberine on neointimal formation,restenosis and the expression of PTEN by western blot methods..Results: Berberine inhibited rat aortic smooth muscle cell proliferation. Berberine improves neointimal formation and restenosis(between group B, groupC and groupA was statistically singnificant, between group B and groupC was statistically singnificant,p<0.05). Berberine upregulation of PTEN expression in a concentration -dependent manner. Optimal concentration of berberine is 200μmol/L for the treat of vascular smooth muscle cell. 14 days before the balloon injury and additional 14days after the balloon injury berberine treat better than a simple Postoperative 14days management for restenosis prevention.Conclusions:Berberine inhibits vascular smooth-muscle-cell proliferation and improves neointimal formation and restenosis after rat carotid artery balloon injury model, probably through upregulation of PTEN expression. |
