| Objective: To observe the effect of probiotics on colon mucosal ulcerative colitis Toll-like receptor 4 and TNF-aAnd the impact of changes in .Methods: Sprague-DawleyRats,which were divided into four group.(1)16 rats as normal control group;(2)16rats as TNBS/ethanol andImmanocomplex group;(3) 16rats as lactic acid bacteria Group;(4)16rats as Bifidobacteria group.First,model group and probiotics group received probiotic for 2 and 4 Weeks.then,us trinitrobenzene sulfonic acid/ethanol andImmune complex rat model induced by ulcerative colon.The rats were killed to get their colonic mucosa and extract the whole cell protein after 24 hours. Rats intestinal mucosa observed changes of Toll-like receptor 4 and TNF-a expression.Results (1)General Normal group after the modeling activities as usual,hair shiny,normal food intake:model rats apathetic,dull hair,often crouched's clothes,food intake decrased,there loose stools,bloody diarrhea,(2)Naked eye Intestinal tissue of normal control group showed a white color,bright and smooth mucosal surface,no congestion,edema.Adhesions between the colon model rats,mucosal congestion and edema in colon wall can be seen.There are several different size ulcer formation.Probiotic intervention group were significantly colonic mucosa edema,and a little ulcer formation(3)HE dyeing:Normal control group see straight glands lined colon,crypt normal,no reduction of goblet cells,and no mucosal erosion,hemorrhage;model rats glands visible part of a large number of colonic damage ,disorder,crypt degeneration,Crypt abscess formation,goblet cells disappeared,a large number of inflammatory cells infiltration.Glands can be seen probiotic intervention group reduced portion crypt abscess,goblet cells decreased inflammatory cell infiltration. (4)Immunohistochemistry TLR4 in each group were expressed within the intestinal mucosa .Normal intestinal mucosa seen in the little brown particles calm;with the normal group,model group glands disappear,we can see many brown granules calm;than the normal group,the probiotic group composed mucosa in the brown particles more.However,less than the model group.Conclusion:From the perspective of the immune system observed by Toll-like receptor 4 probiotics may inhibit NF-kB signaling pathway in order to reduce the expression of inflammatory cytokines TNF-a... |
