Nothapodytes nimmoniana is an important anti-cancer medicinal plant, which possesses varieties of bioactivators, and camptothecin (CPT) is the most important one. In this paper, a determination method on CPT was established for the plant of N. nimmoniana and the CPT content in different parts of N. nimmoniana was detected. Meanwhile, the technologies on extract and purification and HPLC fingerprint of N. nimmoniana were investigated for its effective evaluation of the resources.Firstly, two compounds N. nimmoniana and the HPLC method for the determination of CPT was established. The contents of CPT in different parts of N. nimmoniana were detected. The results indicated that all parts analyzed contained CPT, and the highest content of CPT was 1.87 mg/g from roots,followed by the shoots with content of 1.07 mg/g. Different CPT accumulations were found in different growth phases of stems and leaves. With a view to resource utilization, we concluded that the shoots were the best tissues for exploitation.Several extract processes of CPT in N. nimmoniana were studied by contrast. The Smashing Tissue Extraction (STE) process was finally applied. The best technology of extraction was chosen by the method of orthogonal experimental design. Through repeated solution with solvent and the purification by columnwith purity of above 95% can be obtained.12 samples of N. nimmoniana were analysized through HPLC method with a step gradient of 5mM diammonium hydrogen phosphate-acetonitrile. The HPLC fingerprint chromatogram of N. nimmoniana was established.The inhibitory action on S180 of 9-Me-CPT was investigated. The result showed 9-Me-CPT had a marked inhibitory action on S180; the highest inhibition ratio was 78%.
