Objective This study aimed to investigate the expression and the significance of caspase-1 and cytokine interleukin-1βactivated by caspase-1 after the acute spinal cord injury of the rat.Method 144 adult Sprague-Dawley (SD) male rats are randomly divided into the control group and the spinal cord injury group (n=72 for each group).Two groups are randomly divided into twelve subgroups based on different sampling times after injury. The spinal cord injury group uses the modified Allen's weight dropping method to establish the model of the spinal cord injury. The acute spinal cord injury at the T9 vertebrae level is given contusion spinal cord injury by a weight-drop impactor with a scale from 25mm high in the way of vertical free fall. The control group just undergoes a T9 laminectomy. The rats are sacrificed respectively after injury. The drawn in the center site of injury for the purpose of observing pathological changes of spinal cord by hematoxylin and eosin staining and detecting the expressions of caspase-1 and interleukin- 1 in the method of immunohistochemistry.Result The low expressions of caspase-1 and interleukin-1βare observed in the control group;It is thirty minutes after spinal cord injury that the expression of caspase-1 in two groups is obviously difference. After thirty minutes the expression of caspase-1 in the group of spinal cord injury increases gradually, peaking at 3day after spinal cord injury. After that it decreases gradually, there is not obviously difference about the expression of caspase-1 in two groups at 2W, 3W, 4W;It is thirty minutes after spinal cord injury that the expression of interleukin- 1βin two groups is obviously difference. After thirty minutes the expression of caspase-1 in the group of spinal cord injury increases gradually, peaking at 6 hour after spinal cord injury .After that it decreases gradually, there is not obviously difference about the expression of caspase-1 in two groups at 2W, 3W, 4W.Conclusion There are low expressions of caspase-1 and interleukin-1βin the spinal organization of normal rats;The expressions of caspase-1 and interleukin-1βis significant difference after acute spinal cord injury within 1 week. The increased expressions of caspase-1 and IL-1βmay be involved in the secondary spinal cord injury;The expressions of caspase-1 and interleukin-1βis not significant differences after acute spinal cord injury in 2 to 4 weeks, It means that the low expressions of caspase-1 and IL-1βfor spinal cord organization without biological action.
