Zhangfei is a basic region-leucine zipper(bZIP) protein. It is one of transcription facters participating regulation in endoplasmic reticulum stress and unfolded protein reaction. And it also plays an important role in embryonic development and human disease processes. So far, there are little studies, mainly in the molecular level between Zhangfei and other transcription factors, not in the field of reproductive endocrinology. Therefore, we have prepared its McAb.Expression of Zhangfei in mouse overy and uterus was studied systematically by using immunohistochemical ultrasensitive SP method during the estrous cycle. The results as follows:1 We obtained high purity Zhangfei Fusion Protein by electroeluting, whose concentration was 2.002 mg/mL. The monoclonal antibody to resist Zhangfei can be obtained by culturing hybridoma cell and preparing ascites .Valence of antibody resist Zhangfei Fusion Protein are 1∶2×10~5 in abdominal dropsy and 1:1000 in cell supernatant.2 Zhangfei was presented in the cytoplasm of oocytes during estrous cycle stages.The immunopositive products of Zhangfei was observed in the cytoplasm of the oocyte in the primordial follicles and primary follicles. Zhangfei protein not only expressed in oocytes, granulosa cells also in the expression in the secondary follicles. Zhangfei expressed stronger in granulosa cells in mature follicles. The expression of Zhangfei was increased from proestrus to diestrus. We conclude that it may be related to steroid hormones of ovary which has an important role of regulation of reproductive physiology in the histological changes of ovarian and coordination of follicular maturation.3 Zhangfei expression were seen in uterus during the estrous cycle,mainly in the endometrial luminal epithelium and glandular epithelial cells, almost no expression in other parts. The expression of Zhangfei was lower in estrus while higher in diestrus and proestrus,.It suggests that Zhangfei may be subject to the regulation of ovarian steroid hormones and involve in the regulation of cyclical changes in the uterus. In summary, based on the preparation of the monoclonal antibody to resist Zhangfei, we studied the expression of Zhangfei in the mouse ovary and uterus during the estrous cycle by immunohistochemistry and western blot. All these conclusions provided a good foundation for further study of Zhangfei in the role of the field of reproductive endocrinology .
