Aim To compare the capacity of human umbilical cord blood mesenchymal stem cells and bone marrow-derived mesenchymal stem cells in secreting growth of hematopoietic stem cells in hematopoietic stem cell culture system.And to identify the ability of two kinds to support the hematopoiesis .Methods In this study ELISA method was used for the determination of concentrations of two cell-secreted stem cell growth factor (stem cell factor, SCF), TPO (thrombopoietin, TPO) and interleukin-6 (interleukin-6, IL-6).Results Human umbilical cord blood and bone marrow stromal cells, stromal cells may secrete TPO, SCF and IL-6 and other hematopoietic growth factor. But under the same growth conditions, the concentrations of cell factors secreted by the two kinds of cells were different. The quantity of SCF secreted by Human umbilical cord blood stromal cells (575.7±40.2pg/ml) was higher than that of bone marrow stromal cells(482.2±17.7pg/ml), while that of IL-6 secreted by human umbilical cord blood stromal cells(469.7±27.2pg/ml) is less than the same period in cultured bone marrow stromal cells(935.19±35.4pg/ml); the quantity of TPO secreted by human umbilical cord blood stromal cells (195.5±7.3pg/ml) and bone marrow stromal cells (172.2±9.3pg/ml) were similar.Conclusion Human umbilical cord blood stromal cells and bone marrow stromal cells possess the capability of secreting hematopoietic growth factor, but with varied concentrations.
