Background and Objective:It is very common that influenza virus cause the respiratory infectious disease. It's self-limiting and most of the patients suffering from influenza have light symptom and few complicating diseases. For example,H1N1/A in 2009 is characteristic of widely spread,easily infect,high morality ,which pose a major public health threat for people. As we know, clinicians can diagnose Flu H1N1/A after knowing the patients'medical history, symptom, clinical sign and the result of laboratory examination. However, we'll be able to insulate the patient, control the propagating source and decrease the possibility of spread transmission if we could diagnose influenza virus before it transmits by rapid diagnosing test.During the influenza virul out break,we maybe lost the Flu case if we only consider the patient from clinical sign. Currently, although there are many methods for Flu diagnosis, diagnosing Flu in the early state becomes very important. We shall pay more attention on the patients with clinical sign of flu and take further examination even the preliminary result is negative. Flu virus infect is a limited disease. So we may lighten the symptom and decrease compilation if we use promptly antivirus drugs. But due to the side effects of western medicines and the continuous appearance of drug resistance, traditional Chinese drugs are also an effective way for antivirus.Tianlong compound preparations made by department of traditional chinese medicine of our hospital. There are a wide range of reported used of Tianlong compound preparationin the literature. We can see from historical researches that general flavones possess anti-inflammatory,anti-hypoxia, antianaphylaxis effect function and so on. So our research now is to study for a rapid diagnosing test and tell how to diagnose Flu A quickly in the clinic, evalue the feature of disease and treatment of antivirus. It's also discussed the interaction between general flavones and Flu H1N1/A. PartⅠTo find out a rapid test kit for detection of influenza A virus on the spotObjective:To find out a method of rapid test kit for detection of influenza A in the clinic.Methods:184 nasopharyngeal swabs were acquired from influenza-like patients from November 2009 to August 2010 in First Affiliated Hospital of Guangzhou Medical College of Cheinese Medicine clinic and fever clinic.Among the patients, 96 are men and 88 are women. The average age of the patients is 32.68. All samples were tested by the GICA and DIEFA methods. In addition, qRT- PCR method was used as"gold standard". The sensitivities and the specificities of the two methods in the detection of influenza A virus infection were evaluated and compared.Result:The detection rates of GICA and DIEFA methods were 30% (12/40) and 55% (22/40), respectively. In comparison with the results of qRT- PCR method, the sensitivities and specificities of GICA and DIEFA methods were 42.8%/84.2% and 85.7%/78.9%, respectively. The sensitivity of DIEFA method was higher than the GICA method (p<0.05). The detection rates of DIEFA methods is 45.6%(84/184). In comparison with the results of qRT- PCR method, the sensitivities and specificities of DIEFA methods are 71.5% and 82%. The sensitivity of DIEFA method was not different from real time-PCR(p>0.05).Conclusion:With higher sensitivity, specificity and consistency, DIEFA was a method more suitable for quick detection of influenza A. PartⅡTo evalue the clinical features of influenza patients and the treatment of themObjective: to diagnosis the symptom of influenza infection promptly by age and season, keep good timing of the treatment of antivirus.Methods:We essay the clinic features with influenza infection from history,sign and laboratory examination. Influenza infection patients were randomly divided into three groups:Tamiflu treatment,Tianlong compound preparationin treatment and control group.Result:1.Influenza infection mainly affect youth adults. One patient can infect influenza A(H1N1)virus and seasonal influenza A virus at the same time. Influenza B virus often occurred in May and in June, however, Seasonal Flu A generally occurred in July and in August. Flu A(H1N1)overwhelmed in winter season.2.There were not obvious difference between Flu A(H1N1)and seasonal FluA in white blood cell count(p>0.05).3.Tamiflu treatment and Tianlong compound preparationin treatment could improve symptom of patients, while control group delay recovery(p>0.05).Conclusion:1.The Flu pandemic was relevant to age and season.2.Tianlong compound preparation had some therapeutically effect for Flu virus infection patients. PartⅢTo investigate the effect between flavones and Flu AObjective:To investigate the effect of flavonoids extracted from the Tianlongcompound preparationin against Flu A in vitro.Methods:According to viral replication cycle , three antiviral ways were designed to observe anti–Flu virus effect s of flavone by means of observing cytopathic effect (CPE) and adopting MTT assay.Meantime we use hemagglutination test to know the relationship between flavone and Flu virus.Result:Maximum non-toxic concentration of flavone is 200ug/ml.CPEand HA test display flavone had extent inhibition for FluA.Conclusion:Flavone had extent inhibitory effect s to Flu A virus in MDCK cells.
