BackgroundWith the aging of the human,high-energy trauma and other reasons,a variety of factors have led to increasing incidence of osteoarthritis.But current clinical treatment for osteoarthritis is lack of effective means.With the progress in basic science the research of cytokine treatment for osteoarthritis research has become popular.Transforming growth factorβ1 is a kind of multifunctional cytokines. It can promote variety cell proliferation and synthesis of extracellular matrix components.The studies have shown that TGF-β1 can promote cell cartilage cell proliferation and increase synthesis of cartilage matrix.IL -1 is an important inflammatory factor.It secreted primarily by lymphocytes.IL -1 receptor antagonist as a specific inhibitor of IL -1 can specific binding IL-1 and ihibit it do not express its biological activity.So it can restrain the IL-1 mediated inflammation.This lead to a reducing of cartilage degeneration of osteoarthritis.ObjectiveConstruction rabbit osteoarthritis model by the anterior cruciate ligament transection of the knee.Observation the treatment by vivo interleukin-1 receptor antagonist and transforming growth factor-β1 for the rabbits osteoarthritis.MethodsSixteen three to four month old New Zealand white rabbits were divided randomly into four experimental groups.Group one (normal group) received only opened the left articular cavity.Those in another three groups(IL-1Ra group,TGF-β1 group and control group) received anterior cruciate ligament transection (ACLT).Four weeks later,IL-1Ra group,TGF-β1 group were given vivo interleukin-1 receptor antagonist and transforming growth factor-β1 twice one week and anotheor groups did nonthing.All animals were scarificed eight weeks later after surgery.The samples were stained for tissue slice.ResultsIn general visual study the articula cartilage in IL-1Ra group,TGF-β1 group is close to normal.Control group cartilage is stramineous and can find amotic.Observation by microscope:Chondrocytes of TGF-β1 group proliferation significantly and the morphology of cartilage cells arranged is close to normal articular cartilage.In IL-1Ra group number of cell proliferation is less than TGF-β1 group, the morphology of cartilage cells is disorder compared with TGF-β1 group.These two groups cartilage surface is smooth with no pannus,fibroblasts and fibrous tissue formation. Chondeoucytes in the control group ware significantly reduced and disorganized, morphology changes and can see signs of cell death including nuclear condensation and fragmentation . Cartilage surface roughness, cartilage loss, fracture formation,and this leading to articular cartilage defects.And shows the formation of new cartilage that highlighting the membrane surface. Below the surface of cartilage the pannus formation, fibrosis and fibroblast cell formation.ConclusionBy vivo interleukin-1 receptor antagonist and transforming growth factor-β1 can effectively inhibit degeneration of articular cartilage cell and increase articular cartilage cell regeneration avilably. This will release clinical symptoms so to delay the process of osteoarthritis.
