Objective:To investigate the protective effects of ulinastatin(UTI) on hepatic ischemia-reperfusion injury (HIRI) in rats and the effect of IL-18, to find more effective hepatic-protection drugs and prevention methorats.Methods:Total of 30 Wistar rats were randomly divided into three groups: sham-operated group, ischemia-reperfusion group, ulinastatin injection group. HIRI model was made by Pringle. The activity of SOD, with the content of MDA, the levels of ALTand AST in serum were detected. The activity of SOD, and the content of MDA in hepatic tissue were measured. The rats serum levels of IL-18 and the hepatic homogenate supernatant of IL-18 were detected by ELIS A.Results:Compared with ischemia-reperfusion group:1. the activities of SOD in serum were significantly increased in groups of ulinastatin injection (P<0.05), and the contents of MDA in serum were significantly reduced in groups of ulinastatin injection (P<0.01); 2. ALT and AST levels in serum were significantly reduced in groups of ulinastatin injection (P<0.01); 3. the activities of SOD significantly increased (P<0.05) and the contents of MDA were significantly reduced (P<0.01) in groups of ulinastatin injection in hepatic tissue; 4. The expressions of IL-18 were significantly reduced in groups of ulinastatin injection in serum (P<0.01).Conclusion:1. Ulinastatin injection in HIRI can reduce the injury on the liver caused by lipid peroxidation mediated by oxyradicals, accelerate the elimination of free radicals and has obvious protective effect on the rats of HIRI.2. Ulinastatin injection has the protective effect on the rats of HIRI and the expressions of IL-18 were significantly reduced.
