Objective:To investigate the expression of signal transducer and activator of transcription3 (STAT3) and suppressor of human telomerase reverse transcriptase (hTERT) protein in the retinoblastoma tissue and their relationship with tumor differentiation, invasion and metastasis.Methods:Immunohistochemistry were used to study 54 retinoblastoma tissue samples,the expression of signal transducer and activator of transcription3 (STAT3) and suppressor of human telomerase reverse transcriptase (hTERT) protein in the retinoblastoma tissue were observed,then analyze their relationship with tumor differentiation, invasion and metastasis.Results:(1)In all samples,the positive rates of signal transducer and activator of transcription3 (STAT3) protein in the retinoblastoma tissue were 70.37%(38/54), the expression of signal transducer and activator of transcription3 (STAT3) protein in the retinoblastoma tissue was positively related to tumor differentiation (x2=4.441, P=0.035), optic nerve infiltration(x2=8.347, P=0.004)and clinical stage(x2=7.298, P=0.026); (2) In all samples,the positive rates of suppressor of human telomerase reverse transcriptase (hTERT) protein in the retinoblastoma tissue were 72.22% (39/54), the expression of suppressor of human telomerase reverse transcriptase (hTERT) protein in the retinoblastoma tissue was positively related to tumor differentiation (x2=6.646,P=0.010), optic nerve infiltration (x2=17.244, P=0.000) and clinical stage (x2=9.028,p=0.011); (3) Spearman rank correlation analysis showed:the expression of signal transducer and activator of transcription3 (STAT3) and suppressor of human telomerase reverse transcriptase (hTERT) protein in the retinoblastoma tissue have a significantly positive correlation (r=0.736, P=0.000).Conclusions:(1)The expression of signal transducer and activator of transcription3 (STAT3) and suppressor of human telomerase reverse transcriptase (hTERT) protein in the retinoblastoma tissue were high,and the expression of signal transducer and activator of transcription3 (STAT3) protein in the retinoblastoma tissue was positively related to tumor differentiation, optic nerve infiltration and clinical stage. (2) The expression of signal transducer and activator of transcription3 (STAT3) and suppressor of human telomerase reverse transcriptase (hTERT) protein in the retinoblastoma tissue have a significantly positive correlation, they promoted the invasion and metastasis in the retinoblastoma.(3) The expression of signal transducer and activator of transcription3 (STAT3) and suppressor of human telomerase reverse transcriptase (hTERT) protein can be used as a new markers of malignancy,and help to screen high-risk patients with tumor metastasis. The signal transducer and activator of transcription3 (STAT3) and suppressor of human telomerase reverse transcriptase (hTERT) protein in the retinoblastoma tissue are expected to become the new target molecules in clinical treatment. |
