Objective:To establish the model of adult rat cardiac myocytes in vitro and serum-free culture and to intervene it by using triiodothyronine, then observing that if the triiodothyronine can protect the adult rat cardiac myocytes or not and the relationship with the expression of Fas system.Methods:Establishing a serum-free primary culture method of adult rat cardiac myocytes with the culture medium of M199, observed the cell morphology with light microscope,evaluated the cytoactive and counting the cell after coloured by trypan blue,detected the apoptosis of the adult rat cardiac myocytes with FCM (flow cytometry) and analysed the express of Fas/FasL with Western blotting.Results:The Cell production of the cardiac myocytes is (4-7) x 106 of the each SD rat,and 75-85% of them are active after recalcification.The Cells are rod-shaped, clear transverse striation, good refractive index, sharp edges at both ends; and 5-10% of the rhabdocyte had spontaneous contractions.T3 has the protection to the adult rat cardiac myocytes in vitro and serum-free culture:After 48 hours in vitro and serum-free culture,the myocytes are found that apoptosis rate of the group with T3(13.26±2.46%) was Significantly lower than the matched group(44.32±6.12%) with the analysis of FCM(P<0.05);the express of Fas/FasL of the group with T3 was more subdued than the matched group with the analysis of Western blotting.Conclusion:Apoptosis of the adult rat cardiac myocytes in vitro and serum-free culture could be reduced by using T3, and there may be some relationship with the weaken express of Fas/FasL.
