Objective:To detect the expression of macrophage migration inhibitory factor(MIF),hypoxia inducible factor-1 a (HIF-1 a)in ovarian endometriosis (OEms) and investigate the correlation between MIF and HIF-1 a in endometriosis (Ems).In order to know better the pathogenesy of Ems,and provide theoretical basis to clinical diagnosis and therapy.Methods:Elivision plus Immunohistochemical and Stereology method were used to detect MIF,HIF-1αin 30 cases of ectopic endometrium,30 cases of eutopic endometrium in ovarian endometriosis and 30 cases of endometrium in ovarian teratoma as the normal control.Quantitative images analytical system was performed with computer to analysis the mean optical density (MOD) of MIF,HIF-1αin Stereology.Results:(1) MIF was distributed mainly in the cytoplasm and epicyte of endometrial epithelial cells and mesenchymal cells of OEms. The MOD of MIF in ectopic and eutopic endometrium of OEms and the normal control was respectively (0.180±0.013),(0.158±0.022) and (0.143±0.029),there was statistical significance between groups(F=47.676,P=0.000). The expression of MIF in the different cycle of eutopic endometrium of OEms and normal control endometrial tissue was not significant (t=-0.631,P=0.531;t=1.187,P=0.239),but MIF in proliferative phase of eutopic endometrium of OEms was(0.157±0.018), extremely stronger than that in the same phase of the normal control(0.146±0.029)(t=2.656,P=0.009);and MIF in secretory phase of eutopic endometrium of OEms was(0.160±0.028), obviously higher than that in the same phase of the normal control (0.137±0.030) (t=3.059,P= 0.003). (2) HIF-1αwas distributed mainly in the cytoplasm and nuclei of endometrial epithelial cells and mesenchymal cells of endometrium. The MOD of HIF-1 a in ectopic and eutopic endometrium of OEms and the normal control was respectively (0.077±0.014),(0.070±0.013) and (0.061±0.007), there was statistical significance between groups(F=37.738, P=0.000).HIF-1 a in proliferative phase of eutopic endometrium of OEms was(0.076±0.007), obviously higher than that in the secretory phase(0.059±0.014) (t=6.918, P=0.000),and extremely stronger than that in the same phase of the normal control(0.060±0.007)(t=12.724, P=0.000). (3) Relative analysis revealed that:The correlation coefficient of MIF and HIF-1αin ectopic endometrium of OEms,eutopic endometrium of OEms and the normal control was 0.304(P=0.036),0.123(P=0.295),0.208(P=0.098).Conclusion:(1) MIF and HIF-1αwere found in endometrial epithelial cells and stromal cell, predominantly in glandular epithelial cell.MIF was found in the cytoplasm and epicyte,while HIF-1αwas found in the cytoplasm and nuclei. (2) MIF and HIF-1 a in the ectopic endometrium of OEms was extremely stronger than that in the eutopic endometrium; and in the eutopic endometrium of OEms was obviously higher than that the normal control. (3) The expression of MIF in the different cycle was not significant,but MIF in both proliferative and secretory phase of eutopic endometrium of OEms were higher than that in the same phase of the normal control. (4) HIF-1αin proliferative phase of eutopic endometrium of OEms was obviously higher than that in the secretory phase.HIF-1 a in proliferative phase of eutopic endometrium of OEms was extremely stronger than that in the same phase of the normal control.(5) In the ectopic endometrium of OEms,the expression of MIF has a direct correlation wth the expression of HIF-1α,which means that there maybe have a connection between MIF and HIF-1α,they might be through the different ways of immune regulation, participated in jointly promote angiogenesis, leading to the development of Ems.
