Objective:To investigate the effect of the cold perfusion solution containing Danshen on heme oxygenase-1 (HO-1) expression in rat allogeneic liver transplatation, and protection of donor liver from ischemia-reperfusion injury.and the mechanism of Danshen on donor liver IRI protective function was discussedMethods:A reliable rat model of liver transplantation was established,60 SD male rats were randomly divided into the University of Wisconsin solution (UW) perfusion group (UW group) and the Danshen+UW perfusion group (DS+UW group), ZnPP (HO-1 inhibitor) pretreatment group (ZnPP group),take the blood and liver samples at 24 hours after liver transplantation, and another 10 normal SD rats were used as control group, the same as each surgical group, take the blood and liver samples,serum total bilirubin (TB), alanine aminotransferase(ALT),aspartate aminotransferase(AST) were measured respectively, the liver HO-1mRNA and protein expression were detected by RT-PCR and Western blot respectively,routine pathological section was taken, The histological severity of liver IRI was graded by Suzuki's criteria. And the correlation analysis of HO-1mRNA and its protein expression and liver Suzuki's criteria was taken.Results:①Liver function after liver transplantation in rats:compared with ZnPP group,DS+UW group and UW group TB,ALT,AST significantly lower than ZnPP group (P<0.01),②the expression of HO-1mRNA and protein:the expression of HO-1mRNA and protein was significantly inhibited in the ZnPP group, compared with ZnPP group, the expression of HO-1mRNA and protein in DS+UW group and UW group was more obvious (P<0.05).③liver Suzuki's criteria:DS+UW group (1.4±0.2), compared with ZnPP group(7.3±0.7) (P<0.05), UW group (3.6±0.3),compared with ZnPP group (P<0.05).④The correlation analysis of HO-1mRNA and its protein expression and liver Suzuki's criteria:HO-1mRNA and its protein expression and liver Suzuki's criteria in DS+UW group and UW group was negatively correlated (P<0.01), in the ZnPP group, HO-1mRNA and protein expression and liver Suzuki's criteria was negatively correlated, But the difference was not statistically significant (P> 0.05)Conclusion:DS could upregulate HO-1mRNA and protein expression in rat allogeneic liver transplatation,reduce ischemia-reperfusion injury of donor liver,and produce a protection effect of transplated rat. Danshen in vivo liver perfusion model, the mechanism of protective effects on the liver may be through induction of HO-1 overexpression.
