Neisseria meningitis is a heterotrophic diplococcal bacterium best known for its role in meningitis.With differential capsular polysaccharide antigens,Neisseria meningitis is classified into 13 different serogroups,including A, B, C, D, X, Y, Z,29E, W135, H, I, K, L.With differential outer membrane protein and angarose components, it can be further divided into different serotypes and subtypes. In our country, the most epidemic encephalitis are caused by A groups, others are caused by B and C groups. In recent years, the diseases caused by B groups rose up slowly. A new surface protein, named NspA, which is distinct from the previously described Neisseria meningitides outer membrane proteins was indentified in 1997 Marin. the expression of NspA protein can exist in bacteria in many times. The monoclonal antibody against NspA can protect mice from lethal dose of neisseia meningitis. The combinant NspA vector can vaccine mice, NspA protein is the best potential vaccine with curing epidemic encephalitis.According to NspA sequence in NCBI GENE'S BANK, we design a pair of primer. In this report, we cloned Neisseria surface proteinA(NspA)gene of Neisseria meningitis, constructed PMD-18T-NspA recombinant plasmid and pSIP-409-NspA recombinant plasmid.we construct the pSIP-409 recombinant vector to achieve a high expression of recombined NspA in Escherichia coli.
