This study aimed to investigate the effects of subchronic aluminum(Al)exposure on structure and function of kidney in rats. The model of sub-chronic aluminum exposed of rats was established by different concentration gradient aluminum trichloride in drinking water. 60 Wistar male rats were randomLy divided into 4 groups: control group (C distilled water), low-dose group(L 64.18 mg/kg B.W AlCl3), middle-dose group (M 128.36 mg/kg B.W AlCl3) and high-dose group(H 256.72 mg/kg B.W AlCl3). These rats were sacrificed after given oral administration with different concentrations of aluminum chloride solution for 120 days. Renal pathological changes were observed with Hemotoxylin and Eosin staining (H.E) and Masson staining, and renal lesions were determined by semiquantitative scores. The ultra-structure of the renal tissues was determined by an electron microscope. Blood urea nitrogen (BUN), uric acid (UA), creatinine (Cr), Cystatin C (CysC), concentration of sodion (Na+) in serum, total protein (TP),β2-microglobulin (β2-MG) and creatinine (Cr) in urine were tested by colorimetry, enzyme linked immunosorbent assay(ELISA), latex partical-enhanced immunoassay(LPEIA), ion analysis, urease, immunohistochemical(HI). The content of aluminum in serum, urine and kidney were analysis by graphite furnace atomic absorption spectrophotometry. N-acetyl-B-D-glucosaminidase(NAG) in urine was detected by colorimetry. The results were shown as follows.1 In aluminum exposed groups, the renal tubule was narrowed, the epithelium were dissolved, the renal glomerulus atrophied. A large number of collagen deposition in the renal interstitium and bibrosis obviously. The plasma membrane infolding were swollen, microvillus were swollen and were dissolved. The results proved that aluminum exposed induced the renal injury.2 The contents of CysC,β2-MG, TP and NAG in exposed group were higher than control group (p<0.01), 24h creatinine clearance rate in exposed group were lower than control group (p<0.01), but BUN and UA (p>0.05). The results show that renal function of rats in aluminum exposed was in early stage injury.3 The express of ICAM-1 in kidney of rats in aluminum exposed groups were higher than that in control group, the results showed that aluminum exposed is the one of the important factors of renal injury.4 The contents of Al in serum, urine and kidney of rats in aluminum exposed were higher than control group (p<0.01). The FENa+ in rats of aluminum exposed groups was higher than control group (p<0.01). The results showed that sub-chronic aluminum exposed in rats induced accumulation of aluminum, and enhanced excretion of Na+ in rats.
