Ascorbic acid sesquioxide Ge(IV)liposomes were prepared by calcium acetate gradient method, the methods of detection liposomes content, entrapment efficiency, physical and chemical properties, and stability were established by the experiment.The main results from the experiments, as follows:1. To detect ascorbic acid sesquioxide Ge(IV) of liposomes by UV-VIS soectrophotometry. The ascorbic acid sesquioxide Ge(IV) was good linear relationship in the concentration 40~140μg·mL-1.2. To research the fabrication methods and processing parameters of ascorbic acid sesquioxide Ge(IV) liposomes. The experiment determined which oil-water partition coefficient was best for ascorbic acid sesquioxide Ge(IV). According to the effect of the liposomes encapsulalion efficiency, the experiment was applyed by the five methods of ethanol injection, ether injection, reverse phase evaporation and calcium acetate gradient, finally the liposomes of ascorbic acid sesquioxide Ge(IV) were prepared by the calcium acetate gradient. On the transmission electron microscopy, the liposomes of optimized by orthpgonal design L9(34) were uniform round. The average diameter of the liposomes was 111.6nm and the encapsulation efficiency of the liposomes was 84.12%.3. Preliminary stability results indicated that liposomes of ascorbic acid sesquioxide Ge(IV) were stability at 4℃4. According to the drawing release curve, the liposomes of ascorbic acid sesqioxide Ge(IV) were no burst release phenomenon.The results showed that encapsulalion efficiency of the liposomes which were prepared by calcium acetate gradient method were more high than any other methods. The diameter of the liposomes met expected experimental requirements. In future, the liposomes of ascorbic acid sesqioxide Ge(IV) as a expected new anti-cancer drug would be in vitro experiments, animal experiments and clinical drug experiments.
