Background and Objectives:Premature ovarian failure (POF) is a kind of many factors cause heterologous sexual diseases, refers to appear before the age of 40 women ovarian function decline, amenorrhoea, infertility, and accompanied by Follicle Stimulating Hormone follicle-stimulating Hormone (FSH) leels and Estradiol levels drop. The incidence of POF about 1%~3%, in recent years clinical incidence is increasing trend, and show younger tendency. POF cause fertility loss and low estrogen level has become the two couples healthy life influence big threat. But so far, the etiology and pathogenesis POF not clear, is the reproductive medicine and reproductive health research hot spot and the difficulty. But many scholars think and autoimmune related. Resistance antiovarian antibody (AoAb) is a kind of ovary oocytes, particle within cytoplasm ingredients such as cell for target antigens autoantibody, in immune function when the body to produce abnormal AoAb damage ovary, cause the immune function decline. Someone design rat blood AoAb and animal model T lymphocyte subsets change is autoimmune ovarian premature pathological mechanism of diagnosis and treatment of interpretation, provide new clues. But now AoAb POF to man with blood and T achroacyte subgroup of the changes very little researchers report.This experiment by detecting ovarian premature aging patients serum resistance to ovarian antibodies and peripheral T achroacyte subgroup (CD4+, CD8+ and CD4+ /CD8+ ratio), in order to investigate the changes AoAb and CD4+, CD8+ and CD4+ /CD8+ ratio change in premature ovarian occurrence, development, plays a role more deeply reveal the immune pathogenesis POF, for the early detection and early POF treatment provides the basis.Materials and Methods:In February 2009 to September 2009 in Zhengzhou university second affiliated hospital gynecological clinic patients diagnosed as a group of 20 cases of POF, in another Zhengzhou university two affiliated hospital medical health women of childbearing age 20 cases, age 23 to 39, menstrual cycle, and the age is POF group not statistically significant as control group. Two groups of objects YuQingChen fasting acquisition peripheral respectively two tube, each tube venous serum 2ml, a tube of separation in -20℃save for the determination of AoAb, with ELISA; A tube of EDTA anticoagulation thoroughly incorporated confirm no blood coagulation,20~25℃room temperature preservation, with streaming cells within 6h detection instrument T achroacyte subgroup (CD4+, CD8+ and the percentage of CD4+/CD8 +ratio). Statistical analysis:SPSS 13.0 was used for studies. The data was calculated using mean±SD. The mean of each group was compared by t test treatments. The data was calculated using Pearson correlation analysis.The test level is a=0.05Results:1. AoAb level of serum POF significantly higher than the control group, a statistically significant difference (P< 0.05).2. AoAb positive POF groups of patients significantly higher than AoAb negative antibody levels the antibody level, a statistically significant difference (P<0.05).3. POF groups of patients CD4+ T cells and peripheral healthy women of child-bearing group, difference compared not statistically significant (P> 0.05), CD8+T cells significantly increased and the ratio of CD4+/CD8+ drops apparently, a statistically significant difference (P< 0.05).4. AoAb POF group seropositie patients peripheral blood serum CD4+ T cells and antibody negative CD4+ T cells compared with patients a moderately elevated, difference was statistically significant (P> 0.05), CD8+ T cells significantly increased and the ratio of CD4+/CD8+ drops apparently, a statistically significant difference (P< 0.05).Conclusions:1. In the blood in POF detected with high concentrations of AoAb and high rate AoAb, hints at autoimmune POF growth has played an important role.2. CD8+T cells associated with the immune dysfunction POF closely related.3. The ratio of CD4+/CD8+ decline and the patient appears POF amenorrhea, aging, immune abnormalities related.4. POF patients peripheral T achroacyte subgroup (CD4+, CD8+ and the percentage of CD4+/CD8+ ratio) change and ovarian antigen antibodies appear (AoAb), tip with their own immune related POF.
