| Background And Objective:Liver cancer is one of the most popular malignant tumor in clinic. In the mortality of malignant tumors in the digestive system, it is third. China is a country which has more incidence in liver cancer. The number of suffering cancer every year is 45% of whole world. Because the malignant degree of liver cancer is high, and it develops fast and is difficult to be cured, and its mortality is high, it always is an important project to study in medical field.At present, the treatment methods to liver cancer main includes operation, radiation, drug therapy and biological treatment, and so on. However, total therapeutic effect is not ideal. The discovery of tumor stem cell (or cancer stem cell) makes persons to have new understanding to tumor. People realizes that tumor originates from tumor stem cell. The formation of tumor is a result of the proliferation of tumor stem cell. The quantity of tumor stem cells is very little, and they are only 1% of total cells in tumor tissue. But these cells have drug-resistance and ability to proliferate tumor tissue. For this reason, tumor can be treated when tumor stem cells are wiped out.Tumor stem cells are originally fund in acute myeloid granulocytic leukemia of blood disease, which exist special marker on cellular surfaces. Then, tumor stem cells are also discovered in breast cancer, brain tumor, prostate cancer and lung cancer. There exists some kinds of markers on the surfaces of tumor stem cells. In them, one to be applied widespread is CD133. Some reports show that the cells with CD133 have a close relationship with the formation and development of tumor, invasion and transfer, and drug-resistance. There exists tumor stem cells not only in tumor tissue, but also in tumor cell lines to be cultured in vitro.The hexokinase is an enzyme that can phosphorylate hexose, especial glucose, which is a main compound to get energy by metabolism in most tissues and ogans. Glucose forms 6-phospate glucose by phosphorylation. This is first step of carbohydrate metabolism. Alkaline phosphatase is a kind of enzyme that catalyzes nucleic acid, protein and other molecules to remove phosphate groups. The activities of hexokinase and alkaline phosphatase reflect the abilities of cellular metabolism. Usually, the activities of hexokinase and alkaline phosphatase are high in tumor cells.Soybean flavones and acidic peptide have an inhibition to the proliferation of liver cancer cells. But, it has not been reported whether they can inhibit the expression of CD133 in liver cancer stem cells. So, the influences of soybean flavones and acidic peptide to the proliferation of human liver cancer SMMC7721 and the expression of CD 133 in cancer stem cells have primarily been studied.Materials and methods:1. Cultured liver cancer cell SMMC7721 into logarithmic growth time for using to the following experiments.2. Prepare 100μg/ml,200μ,/ml,300μg/ml,400μg/ml,500μg/ml solutions of soybean flavones and acidic peptide, and treat liver cancer cell SMMC7721 in logarithmic growth time for 24h, 48h and 72h, and detect the rates of living cells by MTT method.3. Treat liver cancer cell SMMC7721 in logarithmic growth time with soybean flavones and acidic peptide, and measure the activities of hexokinase and alkaline phosphatase in the cells by Kit methods.Result:1. After treating liver cancer cell SMMC7721 with soybean flavones and acidic peptide, they show the inhibitions to the proliferations of the cells, and have a close relationship between inhibition and drug concentration.2. When liver cancer cell SMMC7721 are treated with soybean flavones and acidic peptide, they can inhibit the activities of hexokinase and alkaline phosphatase in the cells.3. The results of Western blot show that soybean flavones and acidic peptide can lower the expression of CD133, and depends on drug dosages.Conclusion:1. At experiment concentration, soybean flavones and acidic peptide can inhibit the proliferation of liver cancer cell SMMC7721, and this inhibition is related to the concentration of drug.2. At experiment concentration, soybean flavones and acidic peptide can inhibit the activities of hexokinase and alkaline phosphatase in liver cancer cell SMMC7721.3. At experiment concentration, soybean flavones and acidic peptide can lower the expression of CD133, and shows the dependence of drug dosage. |
