BackgroundRecently, the nature plant essences are currently over the world. Baumann L et al found that some plant extracts could contribute to dermatitis through analyzing a great deal of nature plant compounds. Nayak BS et al reported that the avocado oil rich in plentiful vitamins was delicate enough to be permeable into the skin with softening and hydration effects; moreover, it could accelerate skin damage repair. At present, one kind of vitamin B12 cream containing the avocado oil in prepared in Germany, can be used as topic treatment of psoriasis with safe and long-term characteristics. In our country, the traditional plant drugs with long history are considered important and popular by more and more researchers. Zhong Ch Y indicated that the primary olive oil extracted directly from the fresh olive fruits are rich in Squalane unsaturated fatty acids, lots of vitamins (such as A, D, E, K etc), and anti-oxidant phenols, and the olive oil possesses anti-oxidative, moisture and anti-virus infectious effects on the skin.So far, the information about the biological effects of olive oil and avocado oil on human keratinocytes have been limited. The present study aimed to explore the effects of avocado oil and olive oil on human keratinocyte proliferation and differentiation through investigating the expressions of c-myc and MAPK as proliferative markers, those of filaggrin, involucrin and keratin-10 as differentiation markers and the NF-kB nuclear factor as signaling mediator.ObjectiveTo explore the effects of natural avocado oil and olive oil on human HaCat keratinocyte(KC) proliferation and differentiation.Methods1. The cultured HaCat cells were divided into 3 groups:group A, cocultivated with avocado oil; group O, cocultivated with olive oil and group C, cocultivated with neither avocado oil nor olive oil. MTT assay was used to determine the appropriate doses of avocado oil and olive oil.2. The expressions of c-myc, MAPK, NF-κB, filaggrin, involucrin and keratin10 in each group were examined by immunocytochemistry and immuno-dot-blotting.3. The signal difference between groups was analyzed with Image Analyzer and statistical treatment.Results1. When compared with the grey scaled means (GSM) of respective c-myc, MAPK, NF-κB immunoreactivity (IR) in the control group:101.9±8.9,91.4±5.1,94.3±7.0, the respective GSM in the Avocado oil group:131.4±6.6, 136.3±4.5,134.3±5.2, were increased, P<0.05:2. When compared with the grey scaled means (GSM) of respective c-myc, MAPK, NF-κB immunoreactivity (IR) in the control group:101.9±8.9,91.4±5.1,94.3±7.0, the respective GSM in the Olive oil group:121.1±4.5, 107.9±7.3,106.4±5.4, were also increased, P<0.05. Moreover, in comparison of the respective GSM in the Avocado oil group with that in the Olive oil group, the former was predominant over the latter, P<0.05.3. When compared with the GSM of respective Filaggrin, involucrin, keratin-10 immunoreactivity (IR) in the control group:103.3±7.4,96.1±4.7,99.2± 2.3, the respective GSM in the Avocado oil group:116.7±8.7,109.6±6.5, 105.6±3.4, were increased, P<0.05:4. When compared with the GSM of respective Filaggrin, involucrin, keratin-10 immunoreactivity (IR) in the control group:103.3±7.4,96.1±4.7,99.2±2.3, the respective GSM in the Olive oil group:131.3±9.1,130.9±5.5,129.5±5.8, were also increased, P<0.05. Moreover, in comparison of the respective GSM in the Avocado oil group with that in the Olive oil group, the latter was predominant over the former, P<0.05.5. Furthermore, the data of respective GSM of immuno-dot-blotting in each group were basically coincident with that of immunocytochemistry-IR, there was positive correlation between them in the Avocado oil group, r=0.936, P<0.01; and between them in the Olive oil group, r=0.971, P<0.01.ConclusionAvocado oil and olive oil, can markedly promote KC proliferation and differentiation.
