Panax notoginseng.which belongs to family araliaceae and a herbaceous perennial, is a traditionally chinese herb and has used for medicine purpose for four hundred years.The famous pharmacy scientist of Ming dynasty named Li Shizhen gave this herb a high prise because of its stem,leave and flower have been successfully applied to drug development.Recent years.there are a great number of reports described the bioactivities of saponin aglycones especially focused on their antitumor action. Within these aglycones,PPD as the mainly research target was found to have the action on accelerating the apoptosis process of cancer cells and inhibiting the growth of tumor cells.It shoud to be mentioned that the capability of PPD inducing the apoptosis is better than that of Rg3 which has been developed as a new anticancer agent. Therefore.to find a good technology to obtain PPD and further to use it as the lead compound for the next step to search and prepare another activity novel antitumor drug is meaningful and valuable.In this paper. Orthogonal design was used to optimize the hydrolysis condition and four factors including-solid/liquid ratio.concentration.reaction time and reaction temperature were investigated to study the process of hydrolysis.The primary and secondary influence factors of this process listed as follows:concentration>solid /liquid ratio> temperature> time and the optimization technology of hydrolysis process obtained was:the ratio of liquid to solid material at 1:1 and the concentration of hydrochloric acid with 36.5%,the reaction time and temperature were 1.5 hour and 50℃respectivelyIn this condition, the yield of 20-(R)-PPD.produced from the hydrolysis process of total panax notoginseng by the help of hydrochloric acid,found to up to 8.0% with the purity of 98%.The results shows that this technology is suitable for recrystallization.a easy method to obtain the pure and simple compound, and for the utilization of industry production.
