But the reports concerning about correlation of phosphatase of regenerating liver -3 expression and invasion as well as metastasis in different pathological processes of lung cancer are rarely.Ras homologue C (RhoC) belongs to Ras homologue subfamily of small-molecule G protein superfamily and it is a important molecule in the Rho signal transduction pathway. Many studies have shown that ras homologue C is highly expressed in many tumors and has a highlyrelationship with invasion and metastasis.Objective To investigate the expression of PRL-3 and RhoC proteins and whether they were relative with invasion and metastasis of non-small cell lung cancer(NSCLC); To investigate the function of PRL-3 and RhoC in the migration of A549 cell and the relationship between them, which may provide experimental evidences of the mechanism of PRL-3 and RhoC in tumorgenesis and tumor development.Methods Immunohistochemical staining was used to detect the expression of PRL - 3 and RhoC in NSCLC in 92 cases, and statistical methods were used to analyse statistical significances of its expression in different groups and the relationship between them,The PRL-3 antibody and RhoC antobody were respectively used to block the function of PRL-3 and RhoC; Wound healing assay was used to detect the migration of A549 cell and Reversed transcript-polymerase chain reaction (RT-PCR) was used to detect the expression levels of PRL-3 and RhoC.Results The positive rates of PRL-3 and RhoC expression in NSCLC were 69.6%and 73.9%, and the expression of PRL-3 and RhoC were correlated with TNM stage and lymphatic metastasis and pleural metastasis (P<0.01), and they were correlated with each other(r=0.754, P<0.001). The migration of A549 cell has decreased when the functions of PRL-3 and RhoC were blocked; The expression of RhoC had decreased after PRL-3 was blocked, while the expression of PRL-3 did not change.Conclusion The expression of PRL - 3 and RhoC is higher in the higher TNM stage and lymphatic metastasis and pleural metastasis cases, and closely correlates with each other in NSCLC, which suggests that PRL-3 and RhoC might be in the same signal pathway and PRL-3 might promote the distant metastasis of cancer cell by RhoC and downstream factors.
