Objective:To explore the role of Osteopontin (OPN) in invasive growth and metastasis in animal model of hepatic alveolar echinicoccosis, and observe the effect of exogenous anti-OPN antibody with growth and development of E.multolocularis (Em).Methods:To establish the animal model of Em, mice were infected with 20% Em suspension via liver injection.180 Meriones unguiculatus were divided into 3 groups:Model group:Em inoculation only, Experiment group:Em inoculation and treatment with anti-OPN antibody via tail vein, and Control group:Em inoculation and treatment with blood serum via tail vein. The mice from each group were sacrificed on day 1,20,40,60,80, and 100 respectively. Hepatic echinococcus cyst and metastasis tissue were observed in HE stain, and the expression levels of OPN, MMP2, and TGF-β1 were measured by immuno- staining.Result:, There were no significant difference between Experiment group and Control group, as well as Experiment group and Model group in all time points (P>0.05),while there were significant difference between Experiment group and Control group, and Experiment group and Model group in day 100 group (P<0.05) in the cyst wet weight, and the thoracic lymph nodes metastasis rate. Light microscopy in HE staining showed the pathological change of cyst layer was no significantly difference between experiment group and comtrol group,experiment group and model group on day 1,20,40,60,80, and 100 respectively (P>0.05).The immuno-staining showed that OPN, MMP 2, and TGF-β1 mainly distributed in the germinal layer, the inflammatory cells, and some hepatocytes with different levels. There was no significant difference in the levels of MMP2 between all time points in all groups. The positive expression levels of OPN, and TGF-β1 were not significantly different between Experiment group and other groups on dayl,20, 40,60, and 80 (P>0.05), but the levels in Experiment group were much lower than those in the Control group and the Model group on day 100 (P<0.05)Conlusion:Osteopontin might be involved in the invasive growth and metastasis of hepatic E. multilocularis infection.;exogenous anti-OPN anti-body could inhibit the growth and development of E.multolocularis..
