Objective:in the procedure of extracting DNA from saccus conjunctivae of the patients with atypical acute conjunctivitis and PCR, discuss clinical significance of polymerase chain reaction in the diagnosis of atypical acute conjunctivitis.Method:choose 22 eyes from 22 patients with hyperacute bacterial conjunctivitis,22 eyes from 22 patients with typical acute viral infections,44 eyes from 44 patients with atypical acute conjunctivitis, 22 eyes from healthy person, and take the more severe one from each two eyes. Divide all DNAs into two parts, adding bacteria primer and adenovirus primer separately and carry out PCR method twice, then observe the result in gel imaging analysis after gel electrophoresis. Meanwhile, culture bacteria of the patients with atypical acute conjunctivitis and see the result.Result:1. Groupl (patients with hyperacute bacterial conjunctivitis) has 9 eyes amplified 520bp stripe, positive rate 40.9%; 2 eyes amplified 1004bp stripe, positive rate 9.0%; 1 eye whose stripe possess 520bp and 1004bp, positive rate 4.54%.Group2(patients with typical acute viral infections) has 2 eyes amplified 520bp stripe, positive rate 9%; 8 eyes amplified 1004bp stripe, positive rate 36.3%; 1 eye whose stripe possess 520bp and 1004bp, positive rate 4.54%.Group3(patients with atypical acute conjunctivitis) has 12 eyes amplified 520bp stripe, positive rate 27.27%; 11 eyes amplified 1004bp stripe, positive rate 25%; 3 eyes whose stripe possess 520bp and 1004bp, positive rate 6.81%. According to the result, group1 and group2 has significant difference (X2=5.88,4.66, p<0.05). group1:group3 (X2=2.77,2.29,0.124,p>0.05) and group2:group3 (X2=2.97,1.76,0.124,p>0.05) shows little difference.2. group3 which also procedure bacteria culture method has 17 positive eyes, positive rate 38.6% and this method has little difference from PCR (X2=1.77, P>0.05)Conclusion:1. PCR examination method has clinical practice significance in the diagnosis of atypical acute conjunctivitis.2. Although PCR test positive rate more bacteria culture method, but no obvious difference is not high demand for samples, and time is short, and economy of acute conjunctivitis diagnosis has good application prospect, merits promotion.
