Objective To study PTEN expression in the endothelial cells of sepsisMethods 80 healthy male SD rats were randomly divided into 4 groups (n=20), 25%cecal ligation and puncture group (25%CLP group),50%CLP group,75%group and control group. The rats'survival rate was observed by comparing different CLP rate.50%CLP group is made to sepsis model. Blood and lung tissue samples was taken after sepsis models were established 48 hours. The concentration of blood LPS, IL-1 (3, IL-6 and TNF-a was detected in the septic groups. Detect the PTEN expression by cultivating human umbilical vein endothelial cells, using immunohistochemistry, Westen-blot method, and RT-PCR.Results We found that the mortality of septic rats was increasing, and we concluded that proportion of 50%of cecal ligation was the standard model of CLP. The detection of LPS:the control group was 0.19±0.1 (pg/ml), sepsis group was 3.39±1.26 (pg/ ml); IL-6:control group 20.35±8.11 (pg/ml), sepsis group To 314.81.92±48.7 (pg/ ml); IL-1β:control group 18.73±4.76 (pg/ml), sepsis group was 125.9±17.98 (pg/ ml); TNF-a:the control group was 12.54±5.76 (pg/ml), sepsis group was 105.59±18.97 (pg/ml). In comparison with the control group, indicators were significantly increased in the sepsis group (p<0.05). We also found that in the CLP group the expression of PTEN in the umbilical vein endothelial cells increased significantly by immunohistochemistry, Westen-blot, and RT-PCR (P<0.05).Conclusion The expression of PTEN gene in sepsis HUVEC increased, suggesting PTEN plays a certain role in the endothelial cell injury of sepsis...
