| Objective:Examines the expression of Claudin-1 and Claudin-3 protein in the normal kidney and the renal cell carcinoma tissues, and discusses the carcinogenesis and the clinical significance of Claudin-1 and 3 in the renal cell carcinoma, for finding a new way about the renal cell carcinoma's diagnosis, the treatment and the judgment prognosis.Methods:The specimen about this research includes 61 samples using formalin-fixed and paraffin-embedded tissues from renal cell carcinoma patients after the surgery excised (42 clear cell,13 papillary,6 chromophobe renal cell carcinomas), and uses 10 samples of normal kidney tissues for comparison research. Uses the two step immunohistochemistry to examine the expression situation of Claudin-1 and Claudin-3 in the specimen tissues, then use SPSS 13.0 to analyze relations between its expression and the tumor clinical pathology parameter.Results:Both Claudin-1 and 3 are expressed in all of the normal kidney tissues. In papillary renal cell carcinoma, the high expression of Claudin-1 and Claudin-3 are both significantly differently from in clear cell renal cell carcinoma or in chromophobe renal cell carcinoma. Both Claudin-1 and Claudin-3 expressing in clear cell renal cell carcinoma are related with the tumor pathologicalgrading(the P value respectively is 0.040,0.005). There are not obvious relevance between the expression of Claudin-1 and 3 and pT status in the clear cell renal cell carcinoma or papillary renal cell carcinoma. In addition, the expression situation of Claudin-1 in the cases with lymph node metastasis has a significance statistical difference compared with the cases without lymph node metastasis (P=0.024).Conclusions:Claudin-1 and Clauidn-3 express in the majority papillary renal cell carcinoma, it suggests that Claudin-1 and 3 have an important value of diagnosing papillary renal cell carcinoma. Simultaneously, the expression of Cluadin-1 and 3 was correlation with renal cell carcinoma's pathologicalgrading. And the expression of Clauidn-1 is also related with the lymph node metastasis. It implies that Claudin-1 and 3 may play an important role in tumorigenesis and development, and they may predict the renal cell carcinoma patient's prognosis situation indirectly. |
