Objective:To detect the levels of miR-148a/b and DNA methyltransferase1 protein in pancreatic cancer and paracancerous tissues in order to discuss the function of miR-148a/b and DNMT1 in the tissues of pancreatic cancer initially.Materials and Methods:Collect 30 pairs of Pancreatic cancer tissue samples and paired paracancerous tissue samples from the pancreatic cancer patients who underwent curative pancreectomy. The experiment divides into two parts, the first part uses RT-PCR method to examine the levels of miR-148a/b in pancreatic cancer and paracancerous tissues. The second part uses MaxVisionTM 2 to exam the levels of DNMT1 in pancreatic cancer and paracancerous tissues nearbyResults:1,mir-148a expresses both in pancreatic cancer and paracancerous tissues. The relative expression quantity of mir-148a in pancreatic cancer is 0.425±0.034, and the relative expression quantity of mir-148a in paracancerous tissues is 1.000±0.058, there is remarkable difference, P<0.05.2,mir-148b expresses both in pancreatic cancer and paracancerous tissues. The relative expression quantity of mir-148b in pancreatic cancer is 0.270±0.084, and the relative expression quantity of mir-148b in paracancerous tissues is 1.000±0.076, there is remarkable difference, P<0.05.3. There are 28 examples having high expressions of DNMT1 in 30 examples of pancreatic cancer. The high expression rate is 90.3%; But DNMT1 in the paracancerous tissues has the high 7 expresser, the high expression rate is 23.3%, both comparison has the remarkable difference (P=0.001).conclusions:1.Comparing with paracancerous tissues, mir-148a in the pancreatic cancer is low expression.2.Comparing with paracancerous tissues, mir-148b in the pancreatic cancer is low expression.3. Comparing with paracancerous tissues, DNA methyltransferase1 in the pancreatic cancer is high expression.
