Huaji Koufuye is one of the special species in Jiangxi province, which is make up of Poria (peeled),Endoconcha Sepiae,Endothelium Corneum Gigeriae Galli(stir-baked),Rhizoma Sparganii(processed with vinegar),Rhizoma Curcumae(processed with vinegar),Flos Carthami,Semen Arecae,Omphalia,Fructus Carpesii,Fructus Quisqualis. It can invigorate the spleen to clear retained food, and promote digestion to treat child malnutrition. It has existed in 2005 edition Chinese Pharmacopeia and 2010 edition Chinese Pharmacopeia continuously. the raw specification was vacant on assay, and the identification of Flos Carthami is not stable in the identification item, which was incomplete system and difficult in ensuring the quality. On the base of the raw standard, the new standard inproved Flos Carthami TLC in identification item, and adopted HPLC to determine the contents of p-Coumaric acid in Flos Carthami and Huaji Koufuye.To find out the effective quality control method of Huaji Koufuye, we studied the extraction, separation and structural identification of Flos Carthami. firstly. We have separated one active compound from this plant, which was identified as p-Coumaric acid. Experiments had shown that p-Coumaric acid can restrain Staphylococcus aurei,Shigella dysenteriae,colibacillu,Pseudomonas Aeruginosa at different levels, reduce some side effects coused by anti-cancer drugs and drop blood fat. We have adopted HPLC to determine the contents of p-Coumaric acid in in Flos Carthami and Huaji Koufuye,the method:The separation was performed in a ODS column with a mobile phase of acetonitrile-0.5% phosphoric acid(18:82). The flow rate was 1.0 mL*min-1,and the detecting wavelength was at 308 nm. Results: The linear ranges of p-Coumaric acid were 0.0211~0.4221μg, respectively. The average recoveries were 100.48% and 99.79%, respectively and RSD were 0.6% and 1.8%, respectively. The method is good reproducibility.We have determined the contents of p-Coumaric acid in ten groups of Flos Carthami and 21 groups of Huaji Koufuye, respectively. The method is simple and accurate with good reproducibility, and can be used for the quality control of Huaji Koufuye.
