Fucoidan isolated from sea cucumber(SC-FUC) is a sulfated polysaccharide, and mainly exist in the body wall of sea cucumber. But at present the researches about SC-FUC are very few. The present study investigated the prevention Alcoholic Liver Disease of SC-FUC. In this study, the effects and mechanisms of preventive and therapeutic Alcoholic Liver Disease(ALD).The mice model of acute alcoholism is formed by being affused an overdose of wine, which is used to research the functions of anti-inebriation and liver protection of SC-FUC. The main results of this research are as follows:SC-FUC can effectively put off ebriety time, reduce sober-up time of mice, and enhance the activity of Alcohol dehydrogenase(ADH) and Acetaldehyde dehydrogenase(ALDH) in the liver of mice, which shows that SC-FUC can significantly promote the alcohol metabolism in mice in vivo, improve the ablation rate of alcohol and its metabolic product (aldehyde) to prevent their accumulation and lower harm of liver. SC-FUC have marked functions of avoiding ebriety, anti-inebriation and liver protection. At the same time it can reduce the level of serum alanine aminotransferase(ALT) and aspartate aminotransferase(AST), enhance the activity of antioxidase in liver, which shows that SC-FUC can protect liver by increasing the consumption of peroxy radical from the process of alcohol metabolism and inhibiting lipid peroxidation.To further research the preventive and therapeutic effect on ALD of SC-FUC on the basis of preliminary study, this research adopts a long-term gavage of an overdose of wine to form the alcoholic liver injury mice model, studies the preventive and therapeutic effect on alcoholic liver injury of SC-FUC and has a discussion of its mechanism. The results of this research are as follows: SC-FUC can significantly reduce the level of serum ALT and AST, improve the activity of glutathione–peroxidase(GSH-PX) and the glutathione(GSH) content, reduce the malondialdehyde(MDA) content in liver of mice which are irrigated by wine for a long term, which shows that SC-FUC can improve the antioxidant ability and inhibit lipid peroxidation with the preventive and therapeutic effect on alcoholic liver injury.ALD includes alcoholic fatty liver, alcoholic hepatitis and alcoholic cirrhosis, fatty change is the beginning presentation of ALD. By testing the content of serum total cholesterol(TC) and triglyceride(TG), liver TC and TG, and the activity of ADH and ALDH in liver, this experiment makes a research about the effect of SC-FUC on fat metabolism of the mice which are gavaged by wine for a long term. The results shows that SC-FUC can remarkably reduce the content of fat in both serum and liver, improve the activity of ADH and ALDH. SC-FUC can protect liver by avoiding the accumulation of fat and promoting the metabolism of alcohol. The decrease of liver fat may have a relationship with the functions of SC-FUC, which is able to improve the internal environment of cells, recover tricarboxylic acid cycle, promote gluconeogenesis, reduce the synthesis of fat acids and accelerate its transportation to outsides of cells.The effect of SC-FUC on the expression of CYP450 in the liver of mice which are gavaged by wine for a long term are researched by RT-PCR, further study of the mechanism of the functions of anti-inebriation and liver protection, to provide theories for the application of SC-FUC. The results show that: SC-FUC can increase the expression of CYP2e1 and CYP3a mRNA significantly, which are inducted by alcohol. It hints that SC-FUC is of liver-protecting function by decreasing the expression of CYP2e1 and CYP3a mRNA of liver, the decrease of which maybe related to the functions of SC-FUC to increase the activity of ADH and ALDH, promote the metabolism of alcohol in cytoplast and reduce the alcohol content from MEOS metabolism.In conclusion, the present study provides strong evidence that SC-FUC possesses marked preventive and therapeutic effects on ALD activity by improving the oxidation resistance and regulating MEOS. Researches in this thesis would provide credible theoretical and experimental foundation for the further studies on SC-FUC and contribute to the exploitation and application of sea cucumber.
