AimsThe bronchus asthma, the abbreviation of asthma, which related to many inflammation cells especially mast cells, Eosinophil granulocytes (Eos) and T lymphocytes, was a kind of chronic airway inflammation. Among them, Eos infiltrating was the central section. The pathogenesis of asthma was realated to personal allergic constitution and environmental factors. The pathogenesises involved immune inflammation, neural mechanism, airway hyperresponsiveness and their interactions. Especially, the immune inflammation which included humoral immunity and cellular immunity took part in the pathogenesis of asthma.When the allergens worked on the organism for the first time, T cells could be activated by antigen presentation. IL-4,IL-13, and other cell factors, which were secreted by activated Th cells, worked on B lymphocytes, B lymphocytes could produce nonspecific IgE which could combine to IgE receptor on smast cells and basophilic granulocytes surface. When the same kind of allergens entered the organism again, they could work on mast cells and basophilic granulocytes rapidly, which were activated and secreted histamine, leukotriene, etc. Various cytokines leaded to airway smooth muscle contraction, glandular secretion increased, vascular permeability increased, inflammatory cell infiltration and so on. Meanwhile cytokines secreted by Th cells could affect on Eosinophils, mast cells, monocyte macrophages, aleolar epithelial cells cells. Those cells could also secrete prostaglandin, Eosinophils chemotactic factors, platelet activate factors and so on. Those constituted a complex cytokine network. Interleukin-17 could induce airway epithelial cells and smooth muscle cells expressing various of cytokines, such as interleukin-6, interleukin-8, granulocyte-colony stimulating factor(G-CSF) and neutrophils chemotactic protein-2, etc, they induced various kinds of cells infiltrated in the airway. These inflammatory cells could interacte on each other and could secrete various of cytokines, and all those constituted a complex network.This study established mice animal asthma models through the ovalbumin stimulated to investigate the effects of IL-17 and IL-17 neutralization antibody on Eosnophils and Eotaxin-1 of asthma mice. So we can improve the elabroating of relationship between cytokines in asthma pathogenesis, and provide evidence for the treatment of asthma.MethodsA total of 60 balb/c female mice were randomly divided into five groups: the health group(group A),the control group (group B), the asthma group (group C), the IL-17 neutralization antibody group (group D), and the IL-17 group (group E).Mice in group B,C,D,E were sensitized and challenged with ovalbumin to establish asthma models.The doses of each group's reagent:group C OVA 100μg; group D:OVA 100μg,anti-IL-17antibody 5μg; group E:OVA 100μg, IL-170.1μg, above all were dissolved in 40μl saline (0.9%); group B 40μl saline (0.9%).The pathological characters of lung tissues were observed using HE staining. Eotaxin-1 in mice lung tissue homogenate was determined by ELISA. Group A was not treatmented by particular methods.Results(1) Double forelegs of mice scratched mouth and nose, shortness of breath, abdominal muscle twitched, hair up, incontinence, etc. The biopsy of lung showed that there were various of inflammatory cells infiltrating around bronchis, lung vessels, interstitial tissues of lung, alveolus pulmonis, including of Eos, neutrophils and lymphocytes, etc. The epithelial of airway injuried, the structure disordered; goblet cells in airway were hypertrophy and hyperplasia; More mucus was secreted in the airway, and mucous bloted shaped. It was successful that mice asthma models were established through sensitization and stimulate by ovalbumin.(2)The airway inflammation in the asthmatic mice was characterized mainly by Eosinophils and neutrophils infiltration, which was observed in the normal control group less.(3)The consistency of Eotaxin-1 in lung tissue homogenate were group C, group D,group E,group B,group A from top to ground, representation of x±s,535.86±25.22,461.24±50.79,362.94±35.29,213.32±42.55,above the minimum detection value. The difference between the five groups makes sence.Conclusion(1) We could established asthma models through the ovalbumin aroused for further research.(2) Exogenous IL-17 may inhibit the infiltration of Eosinophils of asthma in mice lung.
