Objective studies indicated that the increase of oxygen free radical and the change of antioxidase activity may drive tumor and tumors usually shows a imbalances of redox state. This suggests that there may be i-nteraction among tumor, oxygen free radicals and antioxidant system. This research compared the level of oxygen free radical, the redox state and the antioxidant ability of Cervical squamous carcinoma and adenocarcinoma cells and investigate the related mechanism.Method①Cervical squamous carcinoma and adenocarcinoma cells were challenged by H2O2, and the proliferation of the cells was observed with the MTT assay;②Cervical squamous carcinoma and adenocarcinoma cells were treated with250,250,800 and 650μmol/LH2O2,nd the cellular ROS, GSH, and GSSG levels and SOD, CAT and GPx activities were determined.Results③Without the treatment with H2O2 Cervical squamous carcinoma cells had the lower ROS and GSH levels and GSH/GSSG ratio and the higher GSSG level and SOD and GPx activities than adenocarcinoma cells. ①In the condition of oxidative stress, the proliferation was inhibited in both types of endometrial cancer cells, especially in type Cervical squamous carcinoma cells.③Under oxidative stress, the GSH level and GSH/GSSG ratio were significantly decreased, and SOD,CAT and GPx activities increased in Cervical squamous carcinoma cell,while the GSH level and GSH/GSSG ratio were slightly decreased and SOD, CAT and GPx activities increased in cervical adenocarcinoma cell.Conclusion①There is lower oxidative stress in Cervical squamous carcinoma cell than in cervical adenocarcinoma cell.②The Cervical squamous carcinoma and adenocarcinoma cells showed different properties in the proliferation after they were challenged by H2O2. The antioxidant ability of cervical squamous carcinoma cell is inferior to that of cervical adenocarcinoma cell, which may be due to the level of ROS increases significantly in cervical squamous carcinoma cellthan adenocarcinoma cell.③In the condition of oxidative stress, the GSH oxidation-reduction system of the Cervical squamous carcinoma cells severely damaged. Although antioxidase system has better response, it can not effectively prevent the increase of ROS and the harm to the GSH oxidation-reduction system, and thus can not very effectively prevent the damage of ROS.④In the condition of oxidative stress,the damage of the cervical adenocarcinoma cell lighter than the Cervical squamous carcinoma cell, and the antioxidase system has better response, which may play a positive role in prevent them from excessive oxidation damage...
