Effect Of Five Traditional Chinese Medicine On Bone Marrow Mesenchymal Stem Cells Proliferation Activity Of Rat In Vitro

OBJECTIVE:①Screening Research on the serum effect of single herb Salvia, Herba Epimedii, Radix Astragali, Saussurea laniceps and Youguiyin on rat bone marrow mesenchymal stem cells proliferative activity in vitro.②To explore the effect of single herb Salvia, Herba Epimedii, Radix Astragali and Youguiyin decoction and ethanolic extract of Saussurea laniceps on rat bone marrow mesenchymal stem cells proliferative activity in vitro.METHODS:①Through high pressure sterilization, Saussurea laniceps after soaked in ethanol and other single herb and compound herb decoction were administered to SD rats by 10 times of the routine dose to do the preparation of herb serum. The bone marrow mesenchymal stem cells (BMSCs) were isolated and cultured from rat femur and humerus by conventional method. The herb serum were administered to the P3 BMSCs with 15%,10%,5% and 1% concentration group. The cells proliferative activity were determined by MTT method after having been cultured 3 days.②The bone marrow mesenchymal stem cells (BMSCs) were isolated and cultured from rat femur and humerus by conventional method. Through high pressure sterilization, The ethanolic extract of Saussurea laniceps and other single herb and compound herb decoction were administered to the P3 BMSCs with10-2,10-3,10-4,10-5 concentration group. The cells proliferative activity were determined by MTT method after having been cultured 3 days.RESULTS:①The herb serum of Saussurea laniceps with all concentration groups promoted BMSCs cell proliferation; the herb serum of Youguiyin promoted BMSCs cell proliferation with 10%,5% and 1% concentration groups (P<0.05).②The herb decoction of Radix Astragali, Youguiyin and the ethanolic extract of Saussurea laniceps promoted BMSCs cell proliferation with all concentration groups (P<0.01)CONCLUSION:①The herb serum of Saussurea laniceps showed the positive effect on rat bone marrow mesenchymal stem cells proliferative activity in vitro.②The ethanolic extract of Saussurea laniceps and the herb decoction of Radix Astragali showed the positive effect on rat bone marrow mesenchymal stem cells proliferative activity in vitro.
