Objective:To investigate the levels of Thl7 cell and Interleukin IL-17 (interleukin-17), IL-21(interleukin-21) in asthmatic children, and study its role in pathogenesis of asthmatic children. To provide theoretical basis and prevention countermeasures for the treatment of asthmatic.Methods:1. In this research,we take Children with asthma (n=66) and healthy controls (n= 48)as the research object. According to the severity of asthma divided the Children with asthma into two groups:the children with acute exacerbation of asthma(n=36), and the alleviatde period asthma children(n=30).2. Separation the PBMC, Adopt flow cytometric profilometry (FCM) detection (CD4+ IL-17+)Thl7 cell percentage.3. The levels of IL-17 and IL-21 in plasma were tested by Enzyme-linked Immuno Sorbent Assay (ELISA). Results:1. Compare with alleviated period asthma children and healthy children, the expression level of Th17,IL-17 were increased in children with acute exacerbation of asthma, and difference was statistically significan (Pa all<0.05), But the expression level of IL-21 was decreased (Pa<0.05).2. The percentage of Th17 cells in PBMC were positively correlated with the level of IL-17 and IL-21 expression level in plasma (r=0.845,0.196;Pa all <0.05).3. The expression leve of IL-17 in plasma was positively correlated with NEU,EOS (r=0.845,0.341,Pa all <0.05).The expression level of IL-21 in plasma was positively correlated with NEU,EOS,LYM (r=0.231,0.296, 0.410,Pa all <0.05).Conclusion:1. The Th17 cells in PBMC and IL-17 common joint participation children asthma systemic immune inflammation.2. IL-17 is expected to become an assist asthma diagnosis new index.And IL-21 may be used as control index of asthma in children.3. Th17 cells may be a two-way regulating cell, its role in the development of childhood asthma need further research.
