| ObjectiveIn this study, the B. hanselae and H.longicornis as subjects, to make a systematic study about the biochemical reaction and survival condition when the suspension was injected into ticks. Making certain that how the defense reaction in the level of invasion of the pathogen, and interadaptation between ticks and bacteria. This study attempts to provide laboratory evidence of vector, and to explore the related tick-borne pathogens to provide clues of immune response mechanism.MethodsThe B. henselae suspension was injected into the hemocoel cavity of partially fed virgin female H. longicornis. Firstly, the death of ticks was observed directly, and then the activity of SOD & CAT and SOD isozyme were assayed at different times after inoculation. Thirdly, the hemolymph's gram stain was also done, the hemolymph and remainder were isolated culture, so that the B. henselae survival of were observed. Finaliiy, the dynamic of B. henselae in tick were observed via dissect ticks. In addition the B. henselae infection was detected from wide ticks.Results1,There are no significant difference between the infected group and control group with mortalty.2,The enzyme activity of SOD and CAT would be inhibited compared to control group in a certain period, though their manifestation were not accordance, there were no difference between two groups at 48h. These illustrate the B. henselae stress was equally to saline solution at 48h, and not huge harm to ticks. There may be different isozymes in ticks of different times and tissues.3,The cluster or single bacteria could be observed under the microscope after inoculated in 96h. But B.henselae had not been found from isolated culture in hemolymph. The bacterial colony was found on the plate of 1h and 24h in four weeks from ticks'remainder. The PCR of salivary gland, midgud, ovary and Malpighian tube, there are almost positive results except very few in 9d, and the different tissue has positive result of every tick at 31d.4,Molecular detection of ticks from wide ticks were detected B.henselae fragment gene from some ticks, and the isolated culture of ticks from Shijiazhuang were obtained.Conclusions1,When the H. longicornis was attacked by B. henselae, the enzyme activity of SOD and CAT would be inhibited in a certain period. There may have different isozymes in ticks of different times and tissues.2,The B. Henselae could survival in hemolymph for some time, and they can enter into other tissues.According to the above laboratory and field survey results, which can prompt Haemaphysalis longicornis would be potential vector of B. henselae. |
