Background and ObjectiveIntracranial tumor is one of the most common diseases in neurosurgery, and brain glioma is the most common, the highest incidence rate, the worst Treatment Effectiveness tumor in intracranial. According for about 35%-50% in constitutional intracranial tumor can happen at any age, the main age is 30-40 years, in addition, people with 10 years are alse common in intracranial tumour. The brain glioma includes astrocytoma, ependymoma, medulloblastoma, glioblastoma and so on The five years survival rate is about 20% in constitutional brain glioma.Compared to other regional tumor, brain glioma is stronger in soakage ability, obscure boundary, higher in recurrence rate, lower in recovery rate, It is one hang-up in neurosurgery, can threaten human heathy seriously. Although there is many kinds of methods including surgery to combine together radiotherapy and chemotherapy for brain glioma, The prognosis is bad still, the radiotherapy and chemotherapy lack specificity for brain glioma, can damage normal brain tissue, cause encephaledema and high intracranial pressure, produce toxic and side-effect in all body, bring about new pains to patient. Because the brain glioma have a malignant biological behaviour for oncogenicity and infiltrative growth, Remaining infiltrative tumor cell is to cause recurrence, the prognosis of patient is not taking on a new look still to remain, it is still a disease which can threaten seriously to health and even vital of human. To search pathogenesis of glioma positively for a effective therapeutic method is always one of the most important focal point research topic in neurosurgery. Along with medical science is to change with every passing day, we know the occurrence and progress of brain glioma deeply.The study discovery:Aggregation and expression of theβ-catenin is related to the tumor's infestation and prognosis;MMP-9 is correlating to infestation of glioma.The purpose of this Experiment is to detect the expression of P-catenin,MMP-9 in brain glioma, investigate the relation and clinical significance of P-catenin,MMP-9 in different grade brain glioma, accordingly to clarity deeply the mechanism of occurrence and progress about brain glioma for providing theory to diagnose and treat and prognosis.MethodsSurgical ablation and preserve well of paraffin-embedded specimens of 68 gliomas were taken from department of neurosurgery in First Affiliated Hospital of Zhengzhou University from July 2007 to July 2010, including 40 males,28 females,the age is 17-62 years old, the averae age is 43.45 years old. All specimens were taken expert appraisal by expert of the First Affiliated Hospital of Zhengzhou University, Pathological grading was classified according to WHO(1999) the standard pathological grades for glioma:low-grade (Ⅰ-Ⅱ) 34 cases, high-grade (Ⅲ-Ⅳ) 34 cases. There are 12 cases normal brain tissue which came from acute craniocerebral injury or cerebral hemorrhage to be cutted as control group. In this experiment,I am making 68 cases brain glioma and 12 cases normal brain tissue adoptjing S-P of immunohistochemisty, detecting the expression ofβ-catenin,MMP-9 in different grade glioma. To contrast the difference of normal brain tissue and low-grade, low-grade and high-grade with Chi square test and Fisher exact probabilistic method, then to analyze the relation ofβ-catenin,MMP-9 with Spearman correlation from ranks.Results1. The expression ofβ-catenin in normal brain tissue and different grade glioma: the expression of P-catenin in normal brain tissue is seldom seen;the masccline in low-grade (I-II) is 52.9%(18/34), for the most part to exist cytoplasm and seldom in intranuclear; the masccline in high-grade (III-IV) is 88.2%(30/34),and 79.4% (27/34) is in intranuclear. The difference of masccline between low-grade and high-grade have statistical significance (P<0.01).2. The expression of MMP-9 in normal brain tissue and different grade glioma: MMP-9 in normal brain tissue has no expression, the masccline in low-grade (I-II) is 47.1%(16/34); the masccline in high-grade (Ⅲ-Ⅳ) is 82.4%(28/34),there are masccline expression both in nucleus and cytoplasm. The difference of masccline between low-grade and high-grade have statistical significance (P<0.05).3. There are significant difference in expression between (3-catenin and MMP-9 in brain glioma, (k=0.376, P<0.05), they are to exist positive correlation.Conclusions1. p-catenin to gather in intra-cellular is to participate the occurrence and progression of brain glioma.2. The high expression of MMP-9 in brain glioma shows high infestation.3. The more of abnormality expression ofβ-catenin,the high of masccline rate of MMP-9,β-catenin decrease the adhesion ability of among tumor cell by negative regulation LEF-1-β-catenin compound.4.β-catenin,MMP-9 is possible to become level of significance of reflecting malignancy level and pathology degree in brain glioma.
