Aim: Myopia is a common eye disease. The cause and pathogenesis of myopia is still not fully understood. The axial length is an important parameter of myopia. Both genetic and environmental factors may contribute to the development of myopia. Recent studies showed that the genetic variants of gap junction protein delta 2 gene (GJD2), insulin-like growth factor-1 gene (IGF-1) and hepatocyte growth factor gene (HGF) are associated with myopia. The aim of the current study is to investigate the possible quantitative association between single nucleotide polymorphisms (SNPs) of GJD2, IGF-1 and HGF and eye axial length or related ocular biometric parameters in Nan'ao middle-aged and elderly people.Method: The subject was the Nan'ao residents >= 45 years old. The axial length and anterior chamber depth, lens thickness, vitreous chamber depth were measured by the A scan. The genotyping of three SNPs, the rs634990 near GJD2, rs6214 in IGF-1 and rs3735520 in HGF were conducted with Taqman assay. The Hardy-Weinberg equilibrium test of genotype was analyzed by Haploview 4.2 statistical software. The relationship between the biometric parameters and the genotypes with different genetic models was accessed by multiple linear regression after control the covariates age and gender.Results: The current study included four hundred and fifty-nine subjects. The associations between the three SNPs and the axial length or anterior chamber depth, vitreous depth were not statistically significant. The SNP rs6214 in IGF-1 was significantly associated with lens thickness in additive model.Conclusions: No evidence of association of GJD2 and IGF-1, HGF with the axial length in our cohort, while IGF-1 could be one of genetic determinants of lens thickness.
