Studies On Chemical Constituents And Pharmacological Activity Of Cynanchum Bengei Decne.

The root of Cynanchum bengei Decne. is a traditional antiaging medicine, with a famous name Taishanbaishouwu. Baishouwu possesses many biological activities: antiaging, antioxidation, anti-inflammation, regulating immunity, which has been well known by many researchers. But many studies on chemical constituents and pharmac- ological activity of C. auriculatum Rogle ex Wight and C. wilfordii(Maxim.) Hemsl, other kinds of plants used as Baishouwu, have been done. However until now only a few works has been done on Taishanbaishouwu. So this paper had studied on chemical constituents and pharmacological activity of Cynanchum bengei Decne..Through the preparation of atmospheric pressure and high pressure liquid chromatography separation techniques, we got nine compounds from Taishanbai shouwu. Structures of them were established, on the basis of spectral data including UV, ESI-MS, HR-ESI-MS, 1D-1~H NMR, 2D-1~H NMR, (13)~C NMR and chemical properties, as follows: bitulinic acid(Ⅰ), baishouwubenzop-henone(Ⅱ), lupeol(Ⅲ), 2,4-dihydroxyacetophenone(Ⅳ), cynandione B(Ⅴ), 2,5-dihydroxyacetophenone(Ⅵ), 2,6,2'-trihydroxy-3-acetyl-4'-(2",6"-dihydroxy-3"-acetyl)phenyl-6'-methyl-benzophe-none (Ⅶ), p-dihydroxybenzene(Ⅷ), 4-hydroxyacetop-henone(Ⅸ). Among all the compounds mentioned above,Ⅶwas a new compound, andⅠ,Ⅲ,Ⅴ,Ⅷwere isolated from the plant for the first time.Antiaging studies demomstrated that the ethanol extract of Taishanbaishouwu as well as the CHCl3 fraction (total steroidal glycoside) had significant antiaging activity. The new compound (Ⅶ) with structure of benzophenone had an antioxidant effect.The result was elvaluated by the method of DPPH. Furthermore, compoudⅦhad a significant effect on inhibiting mycobacterium tuberculosis. In recent years, this kind of compount with the strucure of benzophenone can inhibit HIV-1 reverse transcriptase. So Taishanbaishouwu should have a better prospect for clinical application in the field of tuberculosis and AIDS treatment.
