Objective: To detect the methylation conditions of miR-34a gene in esophageal carcinoma of Hazakh people in XinJiang Province in order to explorer the relationship and significance between esophageal carcinoma and the methylation of miR-34a gene among the population.Methods: Use matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS) analysis to detect the methylation conditions of 59 cases of Hazakh people with esophageal squamous cell carcinomas(ESCC), 32 cases of Hazakh people with non-cancer tissues adjacent to esophageal squamous cell carcinomas(NCAE) and 34 cases of Hazakh people with mucous membrane of normal esophagus(NE); Use software of Cluster 3.0 ,TreeView and GraphPad Prism 5.0 to analysis the distribution of the CpG units among the ESCC,adjacent and NE; Use SPSS17.0 software package to do all the statistical analysis.Results:(1)results showed that the methylation of miR-34a gene in ESCC and NCAE is higher than NE. (2)the methylation rate among ESCC, NCAE and NE, there were 11 units((CpG1.2,CpG3,CpG4,CpG5,CpG6,CpG8.9,CpG14.15.16,CpG17.18,CpG19,CpG20, CpG23)among 15 CpG Units that could be detected, showing statistical significance(p<0.05), and the methylation of miR-34a CpG2,CpG3,CpG4,CpG11,CpG12 in NCAE is higher than ESCC (p<0.05). (3) the methylation rate between different lymph node metastasis , the methylation difference of miR-34a CpG5,CpG8.9 in lymph node metastasis and none lymph node metastasis had statistical significance(p <0.05); the methylation rate between different TNM stage,the methylation of miR-34a CpG8.9 in (â…¢+â…£)is higher than (I+â…¡)stage, showing statistical significance(p <0.05) ; The methylation of miR-34a CpG1.2, CpG6 in HPV16 infection is higher than none HPV16 infection (p <0.05). There were no manifest differences in methylation of miR-34a among gender, age, position, gross, showing no statistical significance (p >0.05).Conclusions: (1) to the population, the methylation of miR-34a gene in Esophageal squamous cell carcinomas and non-cancer tissues adjacent to tumor is higher than normal control, indicating that mthylation of miR-34a gene may be involved in the pathogenesis of esophageal carcinoma. the methylation of miR-34a CpG2,CpG3,CpG4,CpG11,CpG12 in NCAE is higher than ESCC, showing that methylation alteration of these CpGs maybe early-event in the pathogenesis of esophageal carcinoma.(2)Hypermethylation of miR-34a CpG5,CpG8.9 might be correlated with lymph node metastasis of esophageal carcinoma; Hypermethylation of CpG8.9 might be correlated with esophageal carcinoma clinical staging, indicating that it might be involved in tumor progress; Hypermethylation of miR-34a CpG1.2,CpG6 might be correlated with HPV16 infection, and may be involved in the pathogenesis of esophageal carcinoma.
